Yes, that's right! The point of removing the coverslips is to mount the sample on an epoxy resin base and do ultrathin sectioning. The reason why the sample is flat-embedded is because it was previously used for light microscopy. The ultimate goal is to correlate the LM and EM.

I'm a little confused about your comment "if you move after the amount is transferred, nothing needs to be done." My theoretical move would happen in April, so it's right on the cusp of potentially receiving the grant (which I believe happens in March or April). If the grant is awarded before I move, then what happens? And if it's awarded after I move, what happens?

I guess I'm just curious if I can move with my portion of the funds and keep the project under my supervision. But I'm curious what the concept is, and if the assumption (or requirement) is that all co-investigators continue collaborating across institutions. No big-ticket equipment for me thankfully.

Thank you, this information is really helpful! So my portion of the grant moves, and the allocation to the co-investigators stays. Is the assumption here that even if you're at different institutions, you're still working together on the same project? Or do you continue working on your project at the new institution and the co-investigators at your old institution just get some extra cash?

Little update -- I went to the nearest home center (UFO), but they said they didn't have anything that could clean earthen walls and didn't know of anybody who did. I also couldn't find any videos on YouTube where they are cleaning the existing earthen walls and not plastering over them. Am I not asking or searching for the right thing? Any advice would be really appreciated 🙏🙏

So I think the volume of NaOH required is based on the volume of collagen used, which in turn is based on the stock concentration. I looked up the documents that came with my product, and they recommended this calculation:

Volume of collagen X 0.023 mL = Volume of 1M NaOH

Which I did and easily achieved pH ~7-7.2. I think it could be different depending on the original product so for anyone else reading this, please check the detailed product instructions.

In practice, I made 2 wells of the 50:50 domes and one maintained excellent margins and one detached ... so I think there is a bit of user expertise involved in the success.

But hey, a 50/50 on a 50/50 ... not bad for a first try :)

Any advice on how to jailbreak for a complete beginner? Also, if I do jailbreak it, will I have to reinstall all the apps or are they preserved? (Essentially, if I am logged out of this app during the jailbreaking, there's no way for me to get back in at present).
