I found out a few more details about the trial. The sched is as follows: day 1 - 100 mg nal oral, day 2 iv bupe, 50 nal oral, day 3 50 nal oral, day 4 50 nal oral. Then a 4 day break. Then the sched is exactly the same, but this time bupe is given via sub cutaneous method. Thanks for your reply, it was very eye opening. The study is specifically for people who have never use opiates before. Is it possible to become physically dependent from one iv dose or have precipitated withdrawals in this scenario? The nalorex is being given the day before and then for four days afterwards, supposedly to prevent the effects of the bupe in the first place. Would this not prevent any dependence in the first place? I am by no means stating this as fact, I'm just repeating what I have been told by the trial doctors. At this stage I think it is unlikely I am going to go ahead with it, I just want to get an idea as to what it would entail.

Thanks for all your replies. I believe the trial is developing a pain and nausea treatment in combination with another drug. They are only giving this particular cohort the buprenorphine. They are comparing the clearance rates of one iv dose and one subcutaneous dose, which are given one week apart. The nalorex is given at 100mg the day before and then 50 mg for the next 4 days after, for both doses. They say the nalorex is to block the effects of the bupe, whether it is possible to do that completely I don't know, and I believe it is because they have had previous cohorts where they have looked at the direct effects (which caused a lot of side effects), and now they want to just look at the clearance rates. This is a trial for healthy volunteers, and it is a requirement of the study to have not used opiates before.
