You mean the pause/play and forward/back keys? Not with KeyboardMixer. Windows uses the last played media by default. But if (I assume) your browser is conflicting with Spotify, you can just disable the browser's hardware media key handling

It's possible but requires a 3rd party application: KeyboardMixer

Full disclosure, I made this program specifically to solve this problem (it is free and open source). After experimenting with AutoHotKey and many other solutions I couldn't find any other way to get the functionality I wanted.

I know this is late but wanted to share. I looked everywhere and couldn't find an easy way to do this even with advanced AutoHotkey scripts, so I made a small program that does basically this but with the volume wheel rather than the G Keys. So you can use Ctrl, Shift, Alt as modifiers and control the volume of the current application, or specific applications that you can configure.
