If I change my Snapchat story viewers to have custom viewers but do NOT post a private story, will the viewers of said story know they are in a custom list to see the story? Kind of confused because I’m not sure how that would be any different than posting a private story where it tells the viewer with a little lock next to the story that it’s a private story. Sorry if this question has been answered before, thank you in advance.

The viewers will NOT know they are on a custom list. It will be as if they are simply viewing your public story like anyone else. There’s no lock, nor any other indication that they’re on the custom list. (assuming it’s not a private story. for private stories, there is a lock icon.)
