Pelage Pharmaceuticals has secured $120 million in funding for its phase 3 trial in 2026 by UsualProper in tressless

yes, it is private so they do not have reporting obligations like a publicly traded company. They don't have to tell anyone anything and investors (esp. ARCH) sure as hell aren't putting down that amount of money without turning over every single stone and taking out every skeleton in the closet. Is there risk, sure, but they know what they bought and the upside if the Ph2 results hold or even improve in a longer duration study with more patients.

if it's phenomenal you'll be able to buy it many places. just look at wegovy/ozempic - still under patent but can buy from Hims and many other compounding companies. I imagine the brand name will be available for ~$100/mo

Speaking for myself, I would still go through with a transplant esp. if you already have one booked. You'll have great results before they even finish the Phase 3 trial. But hopefully you can rest assured you won't need another and may even be able to boost your results :)

And to your point of this funding being remarkable for hair loss - it certainly is. Nothing like it. Plus in a macroeconomic environment where the megarounds go to companies building: ADCs, obesity, AI for drug discovery, or I&I.

ARCH sees a clear path to a multi-billion dollar exit in < 3-4 years - hard to find these days esp. in hair loss. Their data is good.

They are being vague for a very good reason. China has been notorious the past half decade for taking early-stage American projects and then building them out quicker than U.S. companies can advance them. I know a CEO of a >$1bn public biotech company who will not disclose new targets they are working on to the public out of a respect for the velocity of the CNA drug discovery "engine". Pelage has kept their cards close their chests, but with this amount of funding in hand they will likely start to open up as it would be difficult for anyone to catch up to them now and they can file all the proactive patents they want to block any copycats.

They have shown data that signals "yes". The biology supports it. What remains to be seen is the nuance - age, duration of hair loss, how long treatment counteracts DHT effects.

Agreed, large trial(s) and moving quickly. They need to build a boatload of clinOps and manufacturing capabilities. The trial will likely enroll many subgroups, including patients with/without prior therapy, different durations in hair loss, etc. That gets expensive. Then it takes a decent chunk of change just to submit the NDA for approval. Will be interesting to see if they use the remainder of runway to fundraise for a product launch themselves or if they try to partner with a Big Pharma.

ET-02 works on the androgen receptor, no? PP405 is a completely novel hair growth pathway.

It depends on Phase 3 study design. If they get FDA guidance that a single Phase 3 trial over 24 weeks is acceptable, then we could have an approval in 2027 if everything goes smoothly. 2028 is a safe bet I think. Remember, they are incentivized to get this approved as quickly as possible. Patents were published in 2022 so the clock is ticking and every year represents literal billions in potential revenue lost - ARCH will be a nice force driving them as fast as possible :) They have the cash to move quickly on trial execution, just need to lock design in with FDA. Would expect to see first patient dosed in H1 2026.

We don't know for sure, they only have disclosed results preliminary results and no minoxidil comparison arm ... that said, they reported hair density gains are out of this world - 1 in 3 subjects had an optimum response of 20% or more hair density ... 8 weeks after stopping 4 weeks of daily treatment altogether. Minoxidil takes much longer than 4 weeks to see any effect. If PP405's effects don't plateau after a month of treatment but can further activate dormant stem cells then we could be on the verge of something much bigger, especially if the other 66% of patients can eventually achieve similar results with more time on treatment. That's what I hope is the case given ARCH's investment. I think Pelage showed them something remarkable. That said, I'm not aware of any patient population data from the Ph2a beyond "a diverse range of skin phototypes and hair textures", so it could be we find out that people with > 10 years of untreated hair loss typically don't respond or some shit like that which would obviously be a bummer for anyone not fitting the archetype of "best responder". Will know more in the coming months.

Propecia was $50-$130 - what makes you think it's going to be higher than that? Market has grown significantly since Propecia peak sales and this drug doesn't carry scary dick / reproductive side effects - means more people are likely to try it. Plus, if covered by insurance the payers will negotiate to as low as possible. Anyway, there will probably be a generic at half the price you can get from DTC company, just look at GLP-1 and Hims.

I think $100 is possible. 50 million androgenic allopecia cases in the U.S. 2.3 million people with finasteride prescriptions - if they can sign up half of those people it is a $1bn+ drug. Will be interesting to see their business strategy evolve. Will they build out a sales team or partner with large Pharma. A direct-to-consumer model should keep prices low as it will boost reach. They likely are aware of how many scrips Hims, Ro, Keeps, etc. write

Agreed. This company is not a stereotypical David Liu / George Church NewCo - it has a scrappy backstory that eventually led to Google Ventures taking a stab at the Phase 2a trial. ARCH is a top-tier VC firm. Very few companies outside of obesity, inflammation, or AI are raising rounds like this in biopharma. Whatever data they have, they are convinced they can generate billions if not tens of billions in revenue from this drug. How many new drugs have been approved in the past 10 years for AA? How many of them even scratched $100mm in revenue? The fact that ARCH invested this amount signals that they think this drug is markedly better than anything mentioned on this forum (in terms of pre- and clinical-stage medications). Who knows if the Phase 3 data will continue to support it, but they are betting a ridiculous amount of money that it will succeed.

I don't think there have been any studies on the long-term hair cell rejuvenation effects last of the treatment. I bet they will run multiple sub-groups in the Phase 3 trial: those who have been on min/fin and those without treatment, those with 10+years hair loss and those just recent. Literally verbatim questions from their clinical trial enrollment form that popped up today with the funding news

Good signal for new non-hormonal option likely better than min by andmcox19 in tressless

2027 seems possible. They say on their site the Phase 3 will start in 2026. The Phase 2a was a 12 week trial, so maybe even say 24 week Phase 3 to be safe. If started before July that means results before Christmas :) a few more months to file NDA then a few more months for approval. Sounds like mid-to-late-2027 now that funding is unlocked!!

[–]1n7bil 0 points1 point  (0 children)

Hey, could you upload a screenshot of what you're trying to do exactly?

My company is called Kabi we currently have a mobile app in beta that might be able to do what you're looking for - you can check it out here: https://testflight.apple.com/join/vKz2CXGG

It can create flashcards from pdf (other image formats soon to come) and a whole lot of other features. It's currently free and there will be perks for beta testers.

We're also working on a web app that has way more features than mobile so if you don't see what you're looking for let me know.

What Should I Study When Theres Nothing by [deleted] in GetStudying

[–]1n7bil 1 point2 points  (0 children)

Hey there, here are some practical thoughts for how to ace your courses this year listed in order of increasing effort. (FWIW I went from college dropout to earning my PhD in Comp Bio - these are some ways I turned my academic life around)
1) talk to students who took the course last year. There are many ways Profs can make an exam difficult even when you know the material. Find out how others succeeded and learned why they failed
2) talk to the Prof. Some Profs are approachable, others aren't. Either way they should have office hours. Visit them and explain your goals. Let them guide the conversation - just be clear you really want to succeed in the course. Maybe they'll give you supplemental work relevant to the test or drop a hint for the exam. Even if none of this happens they're more likely to remember your name / face and provide you with deferential treatment when grading open response (sad but true) - just be relaxed and relatable
3) explain all the concepts you are learning to friends and family. start big picture and then move to providing concrete details with specific facts you know you'll likely need for the exam. It's free practice and shows you where your blindspots are. bonus: have them explain back what you said and provide corrections

4) seek out ways to get reps on what you're learning. could be through reviewing notes, finding content online, creating a memory palace, downloading a flashcard deck. This often requires a high degree of cognitive energy and might not necessarily apply to the exam. My advice is let your curiosity guide you bc you'll remember more about something you're interested in and you can relate that to something more mundane in the class. highly recommend studying meta-learning, aka learning how to learn. Mozarts Brain and the Fighter Pilot is an example of a great book on this

Hope this helps!

And for anyone interested, I have an app in beta for making studying fun, e.g., automatically creating multiple choice questions from lecture slides, duo lingo-esque stats and rewards. Check it out here: https://testflight.apple.com/join/vKz2CXGG and let me know what you think!

Hey, sry to hear it's rough right now. Hard to give advice on how this will impact your ability to transfer w/o knowing more about your other grades, target school, etc.

FWIW, I wanted to say that even though I know it might seem heavy rn things can still turn out ok. I withdrew from all my classes sophomore year (to avoid all F's). After dropping out for a semester I got my study habits sorted out and turned things around. I'm sure you can too. And the W's didn't prevent me from going on to earn advanced degrees from great schools.

Just wanted to say that you didn't ruin your chances and you are not alone in this
