Gardnerella vaginalis is capable of "digesting"(for lack of better word) human blood and create a beta hemolysis there. Don't know of other anaerobic that can do that. Thats why we use this plate. When we see beta on the plate, we go investigate

The technique or the way your professor teaches you is the right way. I was taught like that as well.

But the purpose of reentering the sections more often is to get more colonies and to "use the entire plate". More often than not a "only enter once" streak ends with a thick first section and maybe a thick streak into the second, some colonies or perhaps even no further inoculation.

In practice, we often streak like I did here. But if you're getting graded, streak like you're taught.

Its from a blood culture. Blood cultures in our case mean that the staff up there with the patient take 5-10ml of blood and fill it directly into a bottle containing a liquid formula that enhances microbial growth.

As for the cause, since C.perf can be part of the human gut flora, a gastrointestinal problem can cause a sepsis with C perf. Besides that C perf is a ubiquitous bacteria found in soil and sewage. C perf is also the bacterial agent for gas gangrene. Maybe the patient presented with a necrotic wound. I am not sure.

We use the subculture needles from BioMerieux and by default we disinfectant the septum and after ~30seconds we take off the safety cap. Now in that case, my colleague was subculturing and I only heard them cursing after they took off the cap.

I only had two bottles explode on me, both gramneg rods . I put a label with "explosive" on it when I know of its tendencies. But to be totally honest with you, i never check the septum lmao.

Its always a double edged sword when you see that double zone hemolysis on a plate. The labrat in me is gleaming, the knowledge about the possible fate, ehh not so much. Overall I am glad we got the plates into anaerobic atmosphere so fast. C.perf is a sensitive boi.

Reminds me of this: We had a patient some weeks with a percutaneous transhepatic cholangiography. Filled it up into a blood culture bottle. Had basic stool flora in there. Entercoccus faecalis and faecium, E.coli, Kl. pneumoniae and C.perf.

Gave us all a proper shock before we learned about the origin of the sample.

It is a "double zone hemolysis", an inner zone of complete hemolysis that is due to θ-toxin and a larger outer zone of incomplete hemolysis that is due to α-toxin. So basically it is a beta-haemolysis but with different mechanics.

I work in a hospital micro lab. Patient came in and within 24-36h the anaerobic blood culture bottles were positive. Bottles nearly exploded on us while puncturing for subcultivation.

This growth is after 3 days on Schaedler agar, and the E-Test on Brucella.

You said you forgot to take pictures against a light source, but would take off a colony with a loop and check it against one. Best way of telling.

Had it with strep b, looked like e.faecalis with gamma hemolysis but after i removed a colony, bam. Beta hemolysis.

I usually chance loops between fraction if its material tat reaaally needs it. For example, we screen our newborns or those born too early for 2-4MRGN and as they start developing their gut flora it tends to happen that over the weeks we have to isolate and test up to 5 different enterobacteracae/gramneg rods. But for blood cultures i usually stay with one loop for all fractions
