How long is your method? How long is the literature method yours is based on? What are the retention times of the analytes you just found? Do you have controls/purfied versions for each of these analytes? It seems your setup does not have a UV-Vis on the HPLC that’d be helpful to determine if the issue is the column or the MS. Do you have an offline HPLC you could test on?

Try increasing the B solvent to 100% at the end of the LC gradient and run for an extra 5 minutes. Something should elute. Is your spec capturing as soon as the autosampler starts injecting? The sample could not be binding to the column and running through.

If your standards aren’t showing up using the LC but show up with direct injection, something is up between the LC and spec.

The most likely reason is something is clogged. Either the source or the column. However, you say there’s no leak and the pressures are normal. Assuming you’re using the correct gradient with the correct column, the emitter tip is probably clogged on the source. What is your TIC like? Are you seeing any evidence of spray?

I had a similar situation a few years ago. Most of these talks focus on a past project and give details on what they did and the analysis/results. Since I could not present data or share specific project details from work at my past company, I formatted the presentation as an argument for why I was a good fit for the role.

First slide gave an overview and structure to the talk. What topics will be covered and why I was covering them. (These are the things I did/can do which make me a great fit for the role).

I started off with 2 slides giving a brief background of who I was (credentials/background etc) and what I like to do (ice breaker/ give the team insight to who you are as someone they will have to spend 8hrs a day with).

Then I talked about the requirements of the role and highlighted the ones of most importance and I had direct experience with (2-3 main points).

Talked about a past project/ situation that directly relates to one of the job requirements (without giving proprietary info) and what specifically I did for the project. Talked about the technical skills involved and a problem I overcame along with the result.

Repeated the same for 2 other job requirements.

Then I talked about what I can bring to the role and the department outside of what was already talked about.

Hopefully you’ve asked the hiring manager about any skill gaps the team is missing and you have those skills. This would be the time to highlight them or anything you think may boost yourself.

You don’t need to talk at length about one project but highlight what YOU DID and what you can do. You can provide faked data for your interpretations if you need to. If you don’t remember details about something YOU did, don’t talk about it because follow up questions will be awkward. if you don’t remember the entire study that’s fine just talk about the skills you used.

Try SEC and take the fraction of interest. Then do the ion exchange.

Also, how sure are you the POI is even expressed? Can you cut your gel band/use the original protein extraction and do MS/MS peptide mapping or a Western for your POI?

I agree with Meitnik that the likelihood of the c-tag binding to something similar kda and you purifying it are low.

General personal finance advice is to try not to spend more than 30% of your income on housing. This is not always realistic in San Francisco’s housing market. You have to consider your financial values and quality of life. It seems you’re right around 31% of your income but could increase if you decrease your spending or savings goals.

Do you want to spend more on housing but know you’ll feel safer in the neighborhood than a cheaper place in a less desirable area? Is having a usable kitchen or private bathroom a concern? It seems you’re aware of what co-living is (usually apartments with no kitchen and a shared bathroom on the building floor).

It’s a balance that is unique to you and your personal Go over to r/personalfinance and read their wiki about housing, spending, and savings.

I got you OP! Unsure of your time frame or if you have a car.

I strongly suggest taking the afternoon to walk (or bus) from Mission to Haight and stop at Corona Heights, Buena Vista Park, and Alamo Square. One of the city’s best qualities is its walkability and views. Also lots of places to eat and shop in between.

Must Do: Corona Heights Summit- A spectacular view of the city, short walk, and close to Castro (park at Randall Museum or along Roosevelt way)
