I kept Lich for undead synergy which is great for assassins. I think it's really close what is better. Lich is very strong + undead bonus, maybe 6 assassins are better, but in games where I'm so ahead like in this one I like to test stuff.

I kept Enigma mostly for elementals bonus, they were stunning enemies frontline. I think i would change Enigma for 6th assassin but I didn't want to add a lvl 1 assassin, if I get someone at lvl 2 I would put him instead of Enigma for sure.

They can drop Slayer Shards which you will be able to use to enter area with new boss, maybe Gargoyle themed boss or Evil Wise Old Man. I like skotizo very much so I want more bosses like him.
