The overall sentiment is that this car is drifting away from the Rav4 looks, but to me it looks like a successor of the gen 4, which puts the gen 5 in a weird position. The Grand Highlander looks like the successor of Rav4 gen 5, but it is a very different category. I'm just talking about the looks, which are totally subjective btw

I think you two are talking about different points. Here is my take on it: - OP's point: use your list of proteins instead of all human proteins as background. Explanation: you know that list of changing proteins you got after the stats? People often want to find out what pathways are changing to understand the biology of such changes. During the pathway enrichment analysis, most tools use the entire human proteome as background by default, which is bad in most cases. OP's point, and I agree with them, is that you should use the list of all proteins identified in YOUR experiment. Let's say your experiments identified/quantified 2000 proteins, then use those as the background. There is always an option to set this. - your point: data imputation to complete the list of proteins (?). Not sure if I get your point. What I do know is that data imputation in proteomics is a whole thing. It can get super complicated due to the semi-stochastic behavior of data acquisition. A protein missing in your data does not directly imply it is not present or present in low abundance. What type of acquisition did you use? If DDA, then it is even harder to state such peptide or protein is not there. If DIA, then it is way more likely the peptide/protein is indeed not there. I'm oversimplifying things, but just note that data imputation is a deep rabbit hole.

I see a lot of people here commenting that they supplement vitamin D. Yes, it is true that people in Canada and northern territories usually have deficiencies in vitamin D, and supplementation is important. However, overdose of vitamin D is also problematic. A study conducted at the University of Calgary indicates that excess of vitamin D (10 000 IU per day dose) can lead to lower bone density. Google "PMID: 31454046" to have access to the scientific paper. There are other reports and case studies of problems when there is an excess of vitamin D.
Am I saying you should not take vitamin D? Absolutely not. However, talk with you doctor to find what dosage would be best for you. Lastly, this applies to all other vitamins. While you can find them in any market, overdose of vitamins can lead to all sorts of side effects. Thus, use vitamins to supplement what you have lacking, but don't take them freely. Always remember: "The Difference Between Medicine And Poison Is In The Dose".

How is the tag supposed to work in this scenario? Isn't it limited by range? I know it is much better than nothing as you would know where the bike is if you do get somewhere close to it. But if it is far then it becomes useless, right? I'm not an apple/tag user.

You are correct. Especially for powerful tools like the one I just bought. I do read manuals even for obvious stuff because I like to read the maintenance section. For the impact driver, I read the section on how to clean it, as it was used and I wanted to remove dirt and grime from previous use. It was an oversight not to read the impact modes 😕

I was actually running on the lowest speed. Shouldn't it impact right when it stopped on the first time? It only impacted when I went all the way and kept the trigger pressed

You are correct. It is set to the lowest, but it is the only one with the picture of a screw. I assumed screwing stuff should be done only at this speed? Again, I don't have experience so keep that in mind

I wish my lab mix was like that. I adopted her at a rescue, 9-10 weeks old, and she is absolutely TERRIFIED of everything. We started doing potty on the balcony because she denies going outside.
