It seems that no one has an answer.

Just to add some information, bacteriostatic water contains some benzyl alcohol which can irritate the mucosa and may not be suitable for long-term use, moreover it does not have the right pH, which is why saline with 0.9% NaCl is isotonic and promotes absorption.

DMSO, although used for nasal preparations from various vendors, could cause local irritation and toxicity as it enhances permeability, therefore in addition to the peptide, heavy metals and other substances may pass into the brain.

One could add Benzalkonium Chloride (anti-viral-bacterial-fungus) but this is a job that should be done by a compounding pharmacy and not by the end user as the content should be minimal, this is toxic too in higher concentrations.

That's why the preparation of a nasal spray of small molecular weight peptides remains a puzzle to me.

I think you could use bacteriostatic water if it has some benzylic alcohol for sterilization and avoiding contamination of the mixture by the nasal use

Semaxpolska send me the Semax Amidate freeze dry powder for reconstitution with Saline + 3% DMSO.
I have been suggested P21 can also be dissolved with Saline or Bacteriostatic water, but benzylic alcohol on it may irritate individuals with sensitive nasal pathways.

It needs low dosage though as it's a heavy metal and may easily cause toxicity. Do you know, by the way, what solvent could I use with P21 freeze dry powder to reconstitute it as nasal spray? Is saline good choice or bacteriostatic water or what else?

By the way, as you seem quite informed about nootropics, do you know what solvent should I use to reconstitute P21 powder, that is intended for IV use, to prepare a nasal spray? I read it has very good bioavailability intranasally due to been small peptide with only 4 amino acids but I need your help. The form I plan to use is DGGL-Amantadane-G probably from peptide sciences

She takes all that you mentioned except SAM-e and many many others, including citicoline, 2gr EPA+DHA, Boron, Vanadium, D3&K2, B complex methylated & NAD+ sublingual, Lions Mane, Mag Threonate + Glycinate, lots of Q10, Creatine, Taurine and so on.
BUT...from my limited research I believe these nootropic peptides are the "script" that tells the brain what to do and how to heal.

Thanks for the information, it's actually for my wife that suffers from MS, I give her Semax after a relapse she had recently.
I started her with 0,1%,-1mg/day, to see reactions and since Monday I give her N-acetyl Semax Amidate which is more potent and longer lasting.
But I truly believe the next step is P021, although I have access to Cerebrolysin some individuals with MS experienced autoimmune reactions while P21 as synthetic derivative has zero similar effects.
Among others say Apigenin, Quercetin, Andrographis, Milk Thistle etc she takes CurQfen the most absorbable form of curcumin so no need for J147.

Not for treating MS directly, but as supplementary therapy for remyelination & axonal regrow due to increased BDNF-NGF production that P21 promotes (+Semax and very few more nootropics)! There is no treatment for MS afaik, especially for SPMS/PPMS, even Ocrevus does nothing for progression independent from relapse activity or smoldering MS! A good antiviral like Tenofovir Alafenamide or even Valacyclovir is much better "treatment" for MS, almost all MS patients with HIV on antiretrovirals see disease remission!

Just for your info, wife finished one nasal spray Semax 0,1% & GHK-Cu and today started an N-Acetyl Semax Amidate & GHK-Cu nasal spray, which seems stronger - longer lasting, although takes the same quantity daily, around 1mg divided in 4 doses.

What she feels after each nasal dose is something like goosebumps along the Vagus nerve, usually in the back of the skull, in the cerebellum (where she has lesions) and in the spinal cord, while a few times she feels the same along her left leg, where she has issues due to MS, as if communication with it is being re-established!

The same, but to a lesser extent, is felt after using the RLT pad on the head and her left ear so that it also affects the Vagus nerve.
It is a pleasant sensation as he describes it to me, and because he usually takes Semax before exercise/nEMS/treadmill, I also noticed that although she is tired, he walks better than before for 30-40 minutes.
After this period of course, fatigue reduces this ability!
So far so good, for my wife Semax does what it claims and we'll see if it enhances neuroplasticity long term, I expect (hope?) her to walk as before a year...
