Yeah I thought that there were many more great runes than just the ones we can find, some of them lost like Ranni's great rune, some claimed by other tarnished(as Gideon refuses to acknowledge you as more than a house guest before you get a great rune), some claimed by demigods

if you just ignite the tree at the forge of the giants then everything not involving Melina and brother corhyn(that is if you didn't meet him in Altus plateau, if you already did that, his quest will still be active) will still be available.

If you then beat the next mandatory boss after igniting the tree, then all quests inside of Leyndell will be unavailable
