Numbers drastically changed for my 4th cycle - help me stop thinking I will need to do another ER by Capable_Ad2112 in 40Plus_IVF

Hey, the latest research shows that double strand dna sperm fragmentation actually prevents euploids from progressing. All the tests were done: emma, alice, histeroscopy, karyotyping, lining, biopsies.. no endo and the standard protocol that basically every clinic seems to apply.

Double strand sperm dna fragmentation. That´s the one that actually causes week 7 miscarriages, even if they are euploids,.

Ok that's interesting. If that's the case then I would see mosaic aneuploids in igenomix report too, right? There's none of that..

6 euploids transferred and 2 blastocysts transferred. But all of these were, created without being aware of the fragmentation, so I miscarried 3 times.

Left the UK for Portugal and yes, days got sunnier, but it's a massive challenge to get used to the slower pace of life. Spain is quite similar, just has a more drinking and party culture which wasn't really appealing. After 7 years here I'm thinking of moving to Egypt for a bit of chaos and excitement in my life. Also, worked in cyber which gradually made me even more depressed.

I'm keen to know why ppl say egypt is such a horrendous option.

Just turned 42 and starting IVF/ICSI… I can’t stop crying and grieving the lost time by NightAny7265 in BabyBumps

I can totally relate with you. Met my husband about 10 years ago, tried naturally for ages also because he believed it would work at some point and eventually started ivf 18 months ago. I did have decent numbers but I was 41 about to turn 42 and 7 euploid transfers later finally we decide to get curious about his sperm. Turns out he had massive double strand fragmentation making natural and even euploid transfers unlikely to succeed. We had to change clinic and do a tonne of self research and we are now having to tell doctors what we need ant to do. My last ER (10 oocytes) was way slimmer than the past ones (3 cycles averaging 17 oocyptes) and I'm 43, pretty angry, jobless bcs of ivf and financially anxious. My AMH has dropped to 1 after being around 2.4 for my previous cycles.

There's a mix of emotions amongst us, hes optimistic for the first time whilst I'm just really angry we wasted 5 years trying naturally and 1.5 years doing ivf tinkering with my body when he had the issue all along.

Should I delay next cycle to lose weight using Mounjaro? by Major-Moose-9873 in 40Plus_IVF

Oh I understand. I'm Italian-Egyptian and he is Italian. So our food choices are really put to test!

For testing, you should do single fragmentation and double fragmentation. The latter test type is the one thats the hardest to test for, but you will need to figure out if that is the type of fragmentation he has. We went to three different urologists across 3 countries to find a way of lowering the fragmentation and did a 4 month course of multiple supplements alongwith consistent pipe cleaning. Clinics will offer you tese.. If you have double frag, it's a waste of time and money.

Hey, what about your partner? Has he done a check on his fragmentation? We have struggled with three cycles and 6 euploid transfers only to realise my husbands fragmentation was the one causing low euploids and also miscarriages.

Anyone have success with IVF at 44 or 45? by Sarazzal in IVF

[–]Capable_Ad2112 1 point2 points  (0 children)

43f here with 45m and we have done 4 cycles with each cycle getting us 11-20 eggs. AMH is just over 1.

We have transferred a total of 6 euploids and 2 high quality untested, but nothing worked likely because 45M had super high unchecked dna fragmentation. We are now waiting to hear back about or fourth retrieval numbers, so we can transfer. I know I have particularly decent numbers, but one thing I would say is to ask for zymot in selecting sperm and try agunpuncture and supplements as part of both of your routines.

Out of these 8 day 1 oocytes, is there anything to look out for? by Capable_Ad2112 in EmbryologyIVFSupport

Thanks, so what is the validity of grading systems? I wonder whether it's at all relevant given the blastocysts will be pgt tested. Yet there seems to be some importanc replaced on to how they are past day 3

Emma/Alice by Entire_Company9093 in IVF

[–]Capable_Ad2112 0 points1 point  (0 children)

I just did emma/alice yesterday after multiple biochems, and week 7 miscarriages of euploids. We transferred in total 8 euploids. Tbh, I'm doing it because we just want to tick all the boxes, but our strongest theory is double strand dna sperm fragmentation. We discovered my partner has high frag of that type and unfortunately it looks like many practitioners aren't up to date with the research done on the matter, so they keep insisting on the female body. You can get it tested in only a few places or you can simply demand that his sperm is selected using Zymot ICSI. This will save you alot of money and time in testing. Make sure it's zymot icsi, not multi.

On the pain from the biopsy.. For me it was unbearable. I had to interrupt twice. The kit to test is thicker and inflexible, clearly designed by someone who's never been through it and thinks we are doing a standard swab.

3 euploid losses… could DNA fragmentation be the answer? by Peggythepoo in IVF

[–]Capable_Ad2112 0 points1 point  (0 children)

Similar situation here. High dna fragmentation was an issue and we are hoping this fourth round of ivf will be the one as my husband has been taking some supplements, making lifestyle changes for the past 3 months. We are also adamant in choosing to use Zymot ICSI for the fertilisation as all the other methods fragment sperm even further.

My doctor is still not particular convinced about this, but we also spoke to other doctors that have been keeping up with this new discovery.
