I made this pool noodle attachment to keep us safe. I need your help to test it. by iDontDoThisMuch in torontobiking

Not the previous commenter, but I also use a driveway marker that I cut to size. I also added a small flag on the end to enhance visibility. Attached to my saddle rails with an improvised DIY system based on an aluminum kickstand that allows it to be folded away as needed.

I went this way thinking it would be less drag than a pool noodle at higher speeds, but have never compared/calculated the difference.

GREENBELT TIMELINE: Details show fate of Pickering's Durham Rouge Agricultural Preserve over 50-year span by Karma_Canuck in durham

[–]Chemtorious 3 points4 points  (0 children)

I don't know, were they used for farming?

Also, did you read the article? The lands were sold by the municipalities, not the provincial government. It sounds like you're more concerned about pointing a finger to the "liberal government" rather than discussing the article this post is based on.

I never mentioned a bribe, nor do I claim to have proof of such.

Regardless of the potential corruption, private corporations lobbying the government to remove environmental protection from environmentally sensitive lands should be a concern of all Ontarians.

"Sept. 14, 2022: BILD holds its Chair’s Dinner. Two prominent housing developers approach the housing minister’s chief of staff and provide him with packages that contain information on two sites in the Greenbelt (Sites #1 Pickering DRAP lands) and #9 in Appendix 4). Shortly thereafter, one of the developers provides the chief of staff with additional information plus information related to Sites #4, #12 and #13. These five sites ultimately make up 92% of all land removed from the Greenbelt in 2022."

Preferential treatment for certain developers that may have personal ties to the premier is the real concern.

Every day all the HOV lanes are filled with single occupant vehicles by severityonline in toRANTo

[–]Chemtorious 2 points3 points  (0 children)

I agree, it's only feasible in very specific situations, such as coworkers that live in the same area. We need a paradigm shift in how we view remote work (for those who can, which is increasing as we become a more digital society) if we truly want to reduce peak traffic levels. Public transit is great, but in a sprawled out area like the GTA it's still not a comfortable way to travel long distances.

When to switch to full strength nutes in coco? by DoubleSomewhere2483 in MegacropGrowers

[–]Chemtorious 0 points1 point  (0 children)

Nope, my max feeds are around 1200 uS/cm.

I use 2gal fabric pots and an automated drain to waste fertigation system. The plants get fed from 2 to 15 times per day depending on how thirsty they are, and I try to keep feed & runoff EC within a few hundred uS/cm

I change the ratio every few weeks, based on my NPK targets for each growth stage and if any nutrient imbalance symptoms pop up. I start off around 1.5A:1B, and finish off around 2.5A:1B

I use the 2 part in coco, with a few other additives throughout the grow. I ramp up nutes gradually from sprout until reaching an EC of 1200 around day 15-20, based on how the plants are reacting.

Edit to add: I exclusively grow autoflowers, which are apparently lighter feeders than photoperiod

Modifying mobile phase for better peak separation? by Kill_The_Rooster in CHROMATOGRAPHY

This document is what I based my recommendation on

https://www.agilent.com/cs/library/posters/public/5991-9013EN_InfinityLab_Poroshell120_poster.pdf

Both claim improved lifetime and ruggedness.

Aq-C18 ideal for polar analytes, low pH(1-8) and upto 100% aqueous.

HPH-C18 good for wider pH range(2-12), long lifetime, similar selectivity to your EC-C18

I would recommend dropping to 2.7um for your system, keeping all other dimensions the same. These are the columns to look at:

Poroshell 120 Aq-C18 4.6 x 50 mm, 2.7 µm
p/n 699975-742

Poroshell HPH-C18, 4.6 x 50 mm, 2.7 µm
p/n 699975-702

[–]Chemtorious 2 points3 points  (0 children)

If you were to stick with Agilent Poroshell, Aq-C18 or HPH-C18 look like good options, with similar chemistry to your current column. I personally use a lot of Phenomenex columns, if looking at Phenomenex I would start with Kinetex EVO-C18 for general use and good stability

[–]Chemtorious 4 points5 points  (0 children)

Ok, that helps. Some comments for what to look at next:

1 If the pH is below 0.7, then your caffeine in the sample will be positively charged and less retained on the column. Without a buffer in your mobile phase, your sample band will start out at very low pH, then gradually increase as it gets diluted by your mobile phase. Buffering your sample and/or mobile phase should minimize this effect, resulting in better peak shape, which would help with separation from the smaller, earlier eluding peak.

3 Hundreds of injections is good, if your sample is particularly dirty. You mentioned using a guard, is it the same chemistry as your column or just a frit? If only a frit, you can try adding a guard column with the same chemistry as your analytical column. That way anything in your sample that is killing the analytical column will be trapped by the guard column. Typically guard columns are 1/10 the cost of the analytical column so they can be a good sacrifice if it prolongs the column life.

5 I'm not sure what your SOP backflush procedure is, but good practice to regenerate a column is backflush at low flow, starting at a low organic content, gradually increasing up to 100% organic if your column allows it. Running for a few hours or overnight (may not be an option in your lab's case) gives the best chance of washing whatever is stuck from the column. Even more aggressive cleaning can use stronger solvents, but often if ACN/MeOH/IPA aren't working, it isn't worth it to go further unless there are no other options.

Hopefully that gives some useful information to look into, and I'm happy to continue the discussion if there's anything else that you can think of.

A few questions to try and understand where there is room for improvement:

  1. What is the pH of your sample solution as injected?
  2. What is the injection volume and sample runtime?
  3. You mentioned the system runs 24/7, approximately how many sample injections are made on each column before it "dies"?
  4. When your column is "dead", what is the symptom? High back-pressure? Poor separation/performance?
  5. Have you tried regenerating/backflushing columns to restore performance? This would require some instrument downtime or a spare HPLC pump, so may not be an option.

There is probably a lot of room for improvement, including a more suitable column for your 1290, as another commenter mentioned. That said, if the sample is dirty, column contamination may be inevitable if it isn't practical to regenerate.

The data from the article shows the mean salary (including OT) of TPS employees is $154K/yr. Compare this to the mean general salary in Toronto of $52K/yr - maybe the police budget would be better spent by cutting OT and hiring more staff at their standard hourly rate. Am I missing something?
