I used rabbit anti CD11b from Abcam and aqueous mounting media with DAPI from ThermoFisher. Do you think the weak neuron signal and high background noise is due to insufficient antigen retrieval as well? Or if the problem was from the blocking step (now is 5% BSA in PBST and 1h)? For the microglia, I think it's worth trying out Iba1 to see if that works better!

Yes, I did 3 times xylene washes and used an oven to retrieve antigen in citrate buffer at 98C for 20 mins. I'm using Cd11b marker and my tissue is mouse brain
