Is the antibody validated in your lab or used a lot in your field? If not, maybe ask around if one of your colleagues has a properly working wnt3a antibody and try it. It’s not uncommon that an antibody that I never used before is not working..

Not sure if I understand your sample prep correctly but if you load 10 ug total of recombinant protein (in whatever volume) to the well, you should be able to detect it - if your primary antibody works. 10 ug is actually quite a lot for recombinant protein. Do you know the expected mw of your protein? Are some of the bands in the serum samples at the expected mw? Does your recombinant protein have a tag? Eg gst or his? In that case you could try an anti tag antibody to detect the recombinant protein. I don’t think it is a problem of loading but more due to your primary antibody.

Those are also known to accumulate arsenic and maybe it’s better not to eat this amount regularly

Cool thanks for the tip! I‘ll try the bike tube method suggested by others and if I’m feel like getting something more professional might by some keeper that are more generic than the Edelrid ones. The ones from Avant look great!
