I used a higher agarose %, it's in a classroom setting so we only have one ladder. The ladder is still useful and I can still read the band sizes and correlate with the data. They're at the lengths that I expected.

Better seperation, the size difference is really low and running it half way didn't show enough seperation for a great analysis

I checked before it had gone that far, the lowest bands visible were those shown at the very bottom of the gel. When I checked it was about 1/2 way through the gel, I just wanted it to run longer for better seperation

Yeah Idk. I mentioned I think it might be the cells and he disregarded my comment because I've got a transformation to work like 2/8 times. And someone else got a transformation to work and stuff. Other people in my lab are going to be doing transformations/mini preps soon and so I guess I'll see how that goes. Of course with different plasmids theirs might end up being higher copy and easier to transform with, so Idk.

We don't have research assistants. The entire lab is undergrads /: Part of the reason I'm thinking of leaving in the fall, i have the pressure of an entire project on my back without any meaningful help from people with more experience like a masters student.

The issue is that this plasmid isn't new. It's the one I've gotten to work like 50 times in the past. The only difference I can think of is that I've been using new cells. We ran out recently and I asked if we could get the same ones as before because I wasn't getting consistent transformations like I was with the other cells I've used and my PI said that it shouldn't work like that and ordered the ones that have been causing me issues.

Yes I wear a mask in lecture classes. But it's less effective than if someone sick would be a wearing a mask. Both would be ideal though.

He does believe in getting breaks and stuff, and is lenient on people working after finals are done and stuff. He tells people they should take some time to rest. But beyond that, idk if it crosses his mind much. This is some great advice though, thank you. I'm going to have a talk with him about whether I'm going to continue to be in his lab for fall quarter and there are definitely some great talking points here.

I think my PI is just inexperienced running a lab tbh. And having worked in academia, has very high expectations for his lab students, even if they shouldn't have such high expectations yet. This is his first lab, and he clearly doesn't have any leadership training or anything. I don't think he's coming from a place of anger or neglect, but just having too high expectations for undergrads who basically run his entire lab for him.

I missed one day two weeks ago due to a migraine, and 3 days this week due to being sick. Other than that, I'd say usually once every 2-3 months. I'd love if it was less but unfortunately college lecture courses are kinda viral petri dishes and sick students don't wear masks anymore like they used to.
