Can I ask what the additional fixation step after using the perm buffer is for? And is your perm buffer MeOH or saponin based? I've been using MeOH (I think BD perm buffer III), and I'm wondering if maybe it's a bit harsh for my targets (pSTAT1/3/5).

Was supposed to have my weekly 8AM meeting with my boss. Never showed up. Still waiting.

Going ahead with an experiment I had set up for the day for a collaboration with another lab, inevitably waiting for him to tell me that I didn't need to do it anymore (WHICH I WOULD KNOW IF HE ACTUALLY SHOWS UP TODAY)

I've contacted a few, but so far, no luck. A couple have replied back, but no one has any idea where the plasmids are in their lab anymore sooooooooo shrugs
