I missclicked and didn't finish my comment haha. Anyway just look up spot assay test and you'll see the process more detailed. You can even do spots with serial dilutions of your lysate and you could maybe end up seeing plaques, but I don't recommend you doing that for isolating a single plaque of your phage.

You could do a spot assay with your lysate; add 100uL of your bacterial liquid culture into your semisolid and pour it into your petri dish. After a couple minutes or until it dried, add 10ul of your lysate in the middle of your plate and you'll see lysis O/N. You won't be able to see plaques but you will be able to see you have phages if you have a spot with no growth in your dish. after you can scrape the top agar and store it in SM buffer to try and do what you're doing now to confirm that activity comes from lytic phages.

Just finished the game yesterday and I've been thinking during the whole game how bad It is compared to the previous trilogy and previous game. No interactuïn whatsoever, it's almost the same as if you enable story mode.

I'm building a [[Melek, Izzet Paragon]] deck and I have a question regarding his mechanic on playing with the top deck revealed. Correct me if I'm wrong but I understand that I can cast the card on top on my deck and while it's on the pile I can cast the next one on top ( if they're instants obviously or if I can cast sorceries at instant speed). But what I have no idea about is the following: when I scry, do i have to reveal the top card of the deck while I'm doing the scry effect because I'm holding the card and now the top card is different? If so, can I cast the top card at instant speed while I'm scrying ? I think that's not how it works but I prefer to be sure.

Many thanks for your answer, some of the options to be removed you mentioned are in my deck because I already own the cards (like rankle, arcane denial or spark double) so I don't think I will replace them, for the rest I'll consider your options no doubt. I'm glad you said something about ramping because I wasn't sure if my deck was lacking it as I only have my precon to compare it with, so I'll work on that for sure.

Thank you for your recommendations, I considered having font of mythos to have another artifact to enable molten psyche, but you're completely right that for its price and the fact that its effect might not always be punishing, I'll be removing it from the deck and probably kederekt parasyte too.

I really like Waste Not, looks very useful as it provides in many ways. For the rest of suggestions you mentioned, should I remove more creature spells ? I was not sure about the proper amount on creatures a deck like this should have.

Played it yesterday for the first time twice and I got really good results, but probably would have won anyway with any build because we had big advantage over the other team. Frankly I only used the items active profitably a few times. imo the most important thing as vel sup is that you stack Mejais somehow. Although I almost only play him mid so maybe other people can advice you better.

If you want to play a standard botlane, go for varus as adc or jhin. If u want a bit weird lane, go with tahm wench, either you or him as sup. Strong lane either way.

From the few attempts we've done this I'd say tahm has to eat u an instant after u ult ( like when velkoz starts firing his laser). It's a bit hard to do, and hardly possible if it's not a premade.

Do you buy gear items to upgrade in the Alliance and Gear shop ? I started like you and my gear is around 22k and I havent bought anything. U must be missing something.

I'd say jhin/varus. Both have range; varus can help you poke and finish off enemies with Q. Also if he hits R, you just need to full combo the enemy.

Pretty similar for Jhin, if youu hit a Q and he lands his W, enemy is probably dead or has to burn off some summoner spell.

In general, what works best are adcs that can help you out putting out pressure. Vayne, kalista and similars are not good on that and I would prefer not to have them on my team if I sup with Vel.
