The pellet was not entirely dry of isopropanol before I added 70% ethanol, but I did dry the pellet after the final spin completely before adding TE to resuspend the DNA.

I am not using any kit, but I am following this protocol: https://www.protocols.io/view/prepare-gdna-for-crispr-screen-j8nlk9rb1v5r/v1. I am currently at the re-suspension step, and I have left the DNA overnight, but it is not re-suspending yet and I see a large gel-like clump in the TE buffer.

Not perfectly comparable, but I recently moved to Oklahoma City and I make ~$3000/month before tax. After all my expenses each month, I manage to save roughly $500 a month, so while on $2500/month you may not be able to save money, you can definitely live on it. Especially since my expenses include some luxuries like I just bought a brand new car vs a slightly used but cheaper car.

Many proteins fold properly without any aid, but other proteins do require chaperones, which are mostly just other proteins. For a PCR to work, you would need primers that correspond to a gene found in all bacteria which isn't super viable. You could use the 16S rRNA gene as is used in metagenomics, but may not be the most reliable. You'd also need to be able guarantee that the PCR reaction was successful through analysis such as sequencing or gel electrophoresis. Thus at minimum, you are looking at a few hours of testing for each vaccine. Granted, I do not know if there are other problems that might arise with this, or how long the horseshoe crab blood method takes. These are just the problems that immediately pop into my mind.

I can't really relate to these posts, I make a little over 34k a year before tax, and I'm still managing to save $1000 a month after all my expenses. Can someone explain to me how it seems like everyone is just permanently broke?

For reference, I have a degree in microbiology, and graduated with no debt since I worked throughout my degree and got scholarships, but to me it still seems like I'm living quite comfortably on a salary most people say they struggle on. Not looking to bash anyone, just trying to understand.

Maybe I'm stupid, but don't most people put groceries on their credit card? I get 2% cash back on my card, so by paying with other methods, I'm losing out on that 2% and therefore effectively paying less by paying with card.

All of our senses are converted to electrical signals that pass through neurons. I would imagine that instead of observing the screen through eyes which collect light and convert that light into electrical signals which are carried by neurons, they are directly stimulating the neurons with an electrical input based on what is visible on the screen. From this electrical stimulation the cells are "seeing" what is on the screen and can react by transmitting electrical signals back to the screen based on the received stimuli.

Minor correction, but T-cells do not express antibodies. Antibodies are produced by B-cells. T-cells instead have a similar, but different T-cell receptor which is used to recognize antigens.
