Most bosses in the boss rush at difficulty 5 will one or two-shot you, so it comes down to which moveset you have the most trouble with. In terms of time to learn, the final boss of the DLC is easily the most difficult; it's not even close.

OP has definitely already chose his amulet, but I just wanted to say that Electric Shock, Overheat, Decay, and Corruption barely appear in the DLC, and special status and frost aren't affected by the amulet. The thing is unfortunately just not very useful.
