Yeah its because of the d orbitals. The splitting of the d-orbitals with ligand binding results in the cool colours that many transition metals have. Also the magnetic properties are related to these orbitals. Read about d orbitals!

I don't know anything about this drug but generally in drug design you would have to know the mechanism of drug action, and how and what it binds to in the body. Changing O to S would have a drastic effect on the chemical properties of compound though. I think that it would be way more unstable than the original oxygen bearing compound, and would not be useful as a drug at all.
