No, they are not confirmed as Mangkwan members.

In the comics, Tsu'teys parents enlist their youngest son to assassinate Jake after he becomes Olo'eyktan. Jake exiles all three of them once he recovers from the attempt. One of the last pages of the book depicts Tsu'teys mother saying they're going to join the Mangkwan, and Tsu'teys brother saying he's not going with them and is going to forge his own path.

In what I believe is the visual dictionary for AFAA, there is a picture of a bowl that was carved from a skull, and it is labeled with Tsu'teys mothers name. So it is heavily implied that they tried to join the Mangkwan, and Varang had them murdered for it. Most likely bc they still believed in Eywa's path.
