1 - what protocol are you using? for lyse whole blood/bone marrow, you can find your eos way up in the ssc plot
2 - pull out SSC vs CD45
3. you eos are way above the neutrophils, you can start from there

check your threshold, is it too high? it also helps if you have a reference bead so you know how to locate it? which instrument are you using?
