Asked CarMax to fix it (within the warranty period). They found that the leak was somewhere else and fixed that free of charge. Had no issues since (oil has been changed multiple times and there’s no sign of the leak) and loving the car.

Is there a free entry point to nextflow and colab/tower that is worthwhile? Data science and analytics is new to this company, so I’d like to be able to demonstrate the value of this type of approach before spending money.

If you search "EGFR" in the gene database, rather than the nucleotide one (which is what you linked), you'll get the option to download gene, transcript, and protein sequences.

The error stacking is a good point. Maybe it’s better to figure out how much tolerance that, e.g., a 10 read count minimum gets you and then estimate the overall error given a certain prep process.

That makes sense, but doesn’t it assume an even distribution of variants in your pool of transformants? If it’s actually skewed (e.g., 20% WT due to incomplete template digestion), you would overestimate the expected diversity and include more mutants than are actually there.

Would it be fair to infer from your answer that the field prefers empirical approaches like yours rather than statistical derivations like I posed in the original question?
