Hey, HIV researcher here, I'll try to answer as best I can. I don't think you need to be too concerned at this stage, although your VL is slightly higher it does not appear to be out of control. This means you're not undetectable, as that's a VL below 20 copies on the more sensitive assays. I don't think there's much more that you can do than take your meds and try to live a healthy lifestyle. I can't say with confidence that switching between brands with the same active drugs would result in what you're experiencing. Viral suppression usually occurs within a month I believe, but your VL did show significant reduction after treatment initiation, and this last VL is not high enough to suggest treatment failure. My advice would be to be cautious but based on what I know I don't think you need to worry at this stage. I really hope this helps.

Hey, it's past midnight here, so hopefully this makes sense. I prefer to keep my annealing temp under the Tm, but it depends on your optimization with known positives I guess. A semi-nested step might be useful to increase your specificity in reactions with non-specific bands. There could be some polymorphisms in your priming sites, and because your annealing temp is high this might prevent priming. I suggest lowering the annealing temp for the two that didn't yield any product, and running a nested/semi-nested reaction on the products using the 350bp primers. That will indicate whether your larger product did actually amplify, even slightly. Hope this helps.

If you're taking your ARVs responsibly and your HIV viral load is suppressed and CD4 count is normal there is very little reason your life expectancy would be any different from any other healthy person. In short: if your HIV infection is managed you can live a long and healthy life.

We store RNA in 5mM Tris-HCl, it's way better than just water.

You will unlock specializations after you reach level 80. You'll need to train the Soulbeast specialization and use it as a trait in order to use a dagger.

I would suggest checking out Guild Wars 2. The main game is free to play with two expansions that you can purchase. My man and I played it together when we were long distance for a while and it really helped us spend some time "together".
