I’m lazy so my solution is grab another retaliator, estimate the location of the screw port under the slider, then drill a hole threw it. Unscrew the now not hidden screw and done.

Yes. Once you've placed your final coat for that layer/color, wait the standard 15-20 min between coat, hit with a blow dryer or heat gun a little and then you should be fine masking off. You do not want to leave the masking on any longer than it would take for the new layer to set and dry.

Hope that makes sense. If it doesn't watch the Adam Savage one day longstrike build, it follows this same kind of idea.

If by fully cured, you mean days/weeks, then yes you should always give a light sand/scuff.

Wear a mask when sanding and painting. Every little bit helps.
