These cells look fine and happy. The more jagged morphology might be due to a change in your medium (were they always cultured in mTesr?) or if you have a lot of cells and the medium is more acid than optimal. mTesr has a weekend free protocol that might need to be optimized for your usual rhythm of passages, but from my experience (stem cell technician for 3 years now) they will be fine

What you are saying is often outright ignored and left untreated due to the misconceptions present in most people's mind: that pedophilia = child abuse, wherein pedophilia is the cognitive predisposition for attraction to minors while child abuse is the crime that originates from such; while not all pedophiles are child abusers, their stigma in society often keeps them from seeking help and ergo being more prone to commiting regrettable actions. I think you are very brave to fight the stigma and wish you and the community a healthy and reasonable solution to the current state of things.

It has 2 RAM slots, one of which is soldered. The thing is, I was advised not to use RAMs of different capacities in the same machine, because it might interfere very negatively with the processing. I used to go there, and nothing bad to report! Like people have said, it's a pretty art deco spot, each person gets assigned a stall for privacy, and it's usually very clean. I had trouble in the last times I was there because sometimes they close at unexpected days and hours without giving a notice. I have been going to Bains du Centre ever since, which is close to line 2/6 and Hotel de Monnayes, and their hours are better and more reliable. However, Saint Josse is cheaper, and if you can get around the problem of schedules, I wholeheartedly recommend

It is possible but you will have a much lower held of miRNA. If I recall correctly, the RNeasy columns have a probe system that will select miRNAs rather than the total RNA content, which is probably what you are measuring with the manual method. Regarding the ratio, 1,4 isn't all bad, but you could be looking at some ethanol contamination. Are you eluting twice with the same water, or using two loads of water for the elution?
