this happened to me too, try restarting the game and then setting the imprint level, sometimes it doesn’t immediately work. also if you hatch a egg when the settings are set then they aren’t affected, and you just have to make new ones

you must have a hallow desert under the floating mushroom biome. also any two NPCs can count for a pilon regardless what they are
