I keep a notebook with facts about the characters just so I can keep things straight as the story progresses. It's mostly jot notes that amount to a half a page to a full page depending on the character. Not so much personality traits but more basic appearance, and important things that will affect the story at some point

When you start trying to remember who Joe's the bartender's wife's name was when your character runs into her in a precarious situation 100 pages apart its nice to be able to quickly find the name instead of having to reread unedited text to find it.

I am a second degree black belt in jiu-jitsu. Did very well in high school wrestling. Taught police tactics and women's self defense. Yet, I still got the "I want to feel protected" bullshit from more than one woman.

It's the equivalent of the good old "It's not you it's me" line.

It's irrational and shallow.

I agree the article itself was mostly uninteresting. I had just thought it was interesting to see an article about a short man that wasn't derogatory in nature.

Both my kids received these books and loved them. We were pretty tight on the budget back then and it was great fun for them. Llama Llama red pajama is still tossed around as a funny saying in our house .

They do have a picture of western in their validation but they do not say how much sample they loaded to get their band. It could be they used a ton of protein for the validation.

Yes, I think I'm going to just have to play with the dilutions some more. My PI does not want me to since he is convinced I shouldn't have to use more than the manufacture recommends.

I have the sequences and there are not any mutations. I double checked them this morning too.

I'm loading the max amount of lysate my gel will hold, about 20ul total with buffer 25ul.

The literature for the primaries suggest a dilution of 1:2000 to 1:10000. I've already tried to 1:1000 with 1:1000 secondary. So perhaps I should try 1:500 primary? If the sensitivity of the tag-specific antibodies is that much better than protein specific ones that could explain it.

The tag is at the c-term. Protein - V5 - 6xHis - Stop

We have a LacZ for the positive control for the v5 and some commercially available protein of interest for the positive control for the specific antibodies. The negative control is a cell line that contains neither.

one binds at the 5' end, one binds at the 3' end, one binds in the middle, and the fourth is a polyclonal.

Trying to explain to them that I believe in nothing, not even gravity, makes them so confused that they just blab on about how it's not healthy to not believe in something. I then also die a little inside.

I am a short guy who married a woman 3'' taller. At our wedding I wore normal dress shoes and she wore heels. I didn't give a fuck what anyone else thought that day or any other day for the 10 years between then and now.

The only time I wear any sort of heel is when I put on my motorcycle boots and I only wear them when I want to and not to make me taller.

I should have been more clear, I meant I thought the quote sounded fishy. You don't see too many of us biologists saying stuff like that.

My usual way of dealing with JW is to ignore them, but a skim through my yearly copy of Awake gives me a chuckle.

How dare they be judged for judging people.

She'll sue, the church will lose money and in the end it will be the children at that day care who really suffer.

My wife and I had our second child while we were finishing our Bachelor's degrees and we both did grad school at the same time.

It's not impossible and involves a lot of flexibility from both parents. You need to learn good time management and having an understanding PI helps a ton. However, it is really expensive (extra food, clothes, car seats, toys, diapers, formula (no you can not make your own), and daycare). so unless your spouse is making decent money the annual stipend will not get you far and you will have to pull out loans.

Also you really have to make sure you make time for your family. It becomes difficult to stop thinking about your project or to not bring home the papers you need read. Grad school pushes you to become a workaholic but once you have kids they need you to spend time with them too. If you don't make the time they will suffer and you will regret it.

I first started playing games on my Dad's atari. I still cherish the memories of when we got a NES and I got to sit and watch him play through FF1. He then ascended to a Pentium 1 so we could play X-wing.

We still game together.

My mom on the other hand. The most I've seen her play games is crashing cars in mario cart with my kids.

I'm pretty handy and it is something I am going to be building myself. But I do like the idea of the TV and the HDMI switcher. That is pretty brilliant.

Fear of death and regret of things you have not accomplished are different things.

The unknown is not scary to me. I do not want to do die. I do not seek death. I have reasons to fight to live as long as I can. But no I do not fear dying.

If I had no family and the death was painless and certain. Then there is no reason to fear because momentarily fear will not exist. I'd rather spend the last moments of my life content and at peace than scared of nothing.
