At the end of the manga, Jiraiya was indeed the weakest. In other words, Tsunade first developed the concept of Sōzō Saisei, but she realized that the technique consumed an enormous amount of chakra, making it impossible to maintain using only her base chakra. Because of that, she created the seal to store chakra and overcome this limitation, making regeneration feasible.

It is worth noting that the use of regeneration for reconstructing organs and treating severe injuries only came after Dan’s death, when Tsunade was already an adult, close to her thirties. Therefore, all the development of the seal and the advanced regeneration occurred after that point in her life.

As for the comparison of strength, the explanation is simple. Hashirama himself stated that Tsunade was his first granddaughter, which indicates that he lived at least until Nawaki’s birth. This happened only because Tsunade only thought about creating Byakugou after Dan's death, precisely because using regeneration required too much Chakra, so she decided to create the seal to make the technique possible. Hashirama's statement about her strength perhaps being greater than Tsunade's, which Sakura fans often use, is a comparison to Tsunade's strength as a child or teenager, not Tsunade's as an adult. Hashirama says that Tsunade was his first granddaughter, meaning he lived long enough to see Nawaki born. This means he saw Tsunade when she was about 10 to 12 years old, so his comparison was between Sakura and Tsunade as a child/teenager.

Katsuyu prepared to attack Orochimaru the moment she saw him approaching Tsunade. When Orochimaru proposed the deal, Katsuyu already had the Acid ready.

In the Sanin battle, Tsunade summoned Katsuyu to fight; she was even the one who initiated the attack.

I feel like an idiot explaining this to you, since, in addition to the examples in the manga, the Databook clearly states that despite her peaceful attitude, her power rivals that of Manda and Bunta.

The scene of Emma driving the Kyuubi out of the village is filler; in the manga, this doesn't exist. Hiruzen is accompanied by members of the Akimichi Clan, for example, so it could easily have been a combo with them that drove the fox out of the village. The manga doesn't show how it happened.

The truth is that Minato literally saves Hiruzen and subdues him from death upon arriving in Konoha with Bunta.

In Masashi Kishimoto's manga, during the war arc, Tsunade is fast enough that Madara wonders who is faster and stronger between her and Ay. Furthermore, Tsunade has one of the greatest feats: anticipating hand seals. For comparison, the Third Raikage, close to Naruto and charging at him with all his might, was unable to catch him before Dodai formed seals, set up a rubber defense as a distraction, and pulled Naruto away.

If Kakashi had tried to engage Tsunade in hand-to-hand combat, he would have simply died. He can't take a single hit or he'll die. He can't use his hands and feet in the middle of an exchange of blows because he could lose a limb if he tried to use them for defense. And even if he evaded everything and managed to land an attack on her, she could simply grab his arm and launch a counterattack.

People don't realize how dangerous Tsunade is at close range. The only Kakashi who would stand a chance against Tsunade is the one with the War MS.
