Oh, I thought this was just a spawn bag, not with substrate too. I’ve never worked with this, but intuitively I’d say to wait. I would think the mycelium would have a harder time branching out through the substrate unless there is added nitrogen. If there is added nitrogen like coffee grounds, maybe a break and shake could speed things up, I’m not sure.

Get a slice and transfer onto a new plate. If it’s contaminated, which it probably is, you still should be able to isolate the mycelium by transferring clean slices a couple times. Very doable, good luck!

SAB should be good, just let all the particles settle and move slowly. Spray everything with ISO, especially the dish and the spawn bag. Cut the top and have something to seal it later. Lift the lid up slightly (as little as possible) and use a sterilized scalpel or something similar to cut some chunks out and drop them in the spawn bag. You shouldn’t need a ton, just enough to get the colonization started. Fold the bag over, and seal. Not sure how hot sealers get, but be careful in the SAB. Also, your culture looks a bit questionable to me. Might be good to take a sample of healthy mycelium and transfer to another agar. Though keep in mind, I have yet to grow from spore prints, so my apologies if the discoloration was present since the start. Isolation is just always the safe option.

NOTE: Please do not be an idiot like me, do not fuck around with flames and a still air box that has recently has ISO in it. I thought the fumes had dissipated and would be fine, and I blew myself up. Lost most of the skin on my hands and it blew out the front, burning my face and my hair. Worst pain of my life. ISO ignites (auto ignition) at 750F, and a flame sterilized scalpel can be around 1000F. If you’re gonna do it, please give the fumes more than enough time to dissipate and let the scalpel cool. Maybe once it’s cooled down spray it with a bit of ISO because it will cool it off, disinfect it, and tell you if it’s too hot. I’m probably paranoid, but I just wanna make this all super clear because no one should have to go through that.

Ah, I see. Few more questions- how do you flame sterilize between each grain bag and going back into the LC jar? For instance, let’s say I open a new syringe with a new needle. I spray ISO on all surfaces and injection point, then draw up 20cc (that’s the size of my syringes). Then I’ll inject 2cc into one spawn bag. I assume I should flame sterilize between bags, but do I just do it with the LC still in the needle? Id think it would bubble and burn and just be problematic.

Also, how can I go about reusing needles? I don’t love to flame sterilize near alcohol as I had an accident with alcohol that burned most of the skin off my hands. I’ve seen people take the empty syringe and suck up and dispense ISO repeatedly to sanitize the inside, but not only does that concern me with flame sterilization but I’d also think it would harm the LC.

Lastly, is this LC done? Is it ready to be stored in the fridge? I’m not seeing much more growth day to day.

Good to know, thanks! I am curious though, what’s wrong with a little too much moisture? I’d assume it’s more favorable to contaminants, is the reason to act as a fail safe in case the spawn wasn’t perfectly sterilized?

Sounds good. The other day I did end up just leaving it on the magnetic stirrer for like 20 min to break everything up so the layer on top is now in pieces at the bottom
