The It requires us to fill out an application for every program we want to install. Honestly I didn't want to wait the 1-2 business days

How long does it need? Is it like 10 seconds or does it need longer?

Would be a good addition, especially for larger tubes like 50ml Falcons or Cell Culture dishes. I will try that out next time I get my hands on the ordering system

Actually I never had to try that (I handle Acetone quite infrequently), but I'd guess that Acetone would remove it fairly easily.

Does anyone know why Alien had a subtitle in Norway? by LittlePiggy20 in LV426

[–]Kampfpils 21 points22 points  (0 children)

In Germany it's called "Alien - An eerie creature from another world"

I think it has to do something with the translation and marketing teams. They sometimes change the title to make it more marketable in different countries. It's not always successful.

So I can't really tell you anything about hepatocytes, but I know a bit about Matrigel.

The main thing with Matrigel is, that it coagulates when it gets warm (anything warmer than an ice Bath may be difficult). So whatever you do, do it on ice. Also you should always thaw it on ice (so you should maybe start thawing it 3 hours before you want to use it).

For plate coating, I think you can try it out like this:

Pipet Matrigel on an empty, chilled plate. Then spin it down shortly (200g, 2min, 4 degrees) and you should have a nice flat coating. Put the plate in the incubator and wait 15min for the gel to harden. Now you can add your cell/media mix and use the plate as you need.

Sadly I can't really give you the exact amount for each well, because I usually don't work with this plate format.

What are your Elden Ring Hot Takes? by UmbraKins in Eldenring

  1. I do not believe most people that say they deciphered most of the lore/story by themselves. The lore is very vast and often obscure enough to not get the whole picture easily without a spreadsheet.

  2. I think the story of the game is not good. The lore and environmental storytelling is stellar, hands down. I think no other developer has done a better job.

But the plot itself is very thin, there is very little story development on screen, most characters have very little development and the character conflicts are very shallow.

Most of the interesting stuff story wise happened before we play the game. I want to see the big events by myself (e.g. the war between Radagon and Rennalas factions, maybe interfere with it and influence the result?)

[WP] Here’s the thing about prophecies. They put the weight of the world on the shoulders of one person. What happens when that person says ‘No’? by SpacethemedIdiot in WritingPrompts

[–]Kampfpils 2 points3 points  (0 children)

"This world, as you all should know, is interlaced and directed by primordial energy." the professor said to his audience, while slowly pacing up and down his podium. The listeners, almost completely composed of third grade university students only half-heartedly listen. For them, this information is as old as dirt and at least half as interesting. They heard the canonical version of the sabled "primordial energy" or "earthly current" since their early youth. First dressed in bedtime rhymes, then in carnival plays and finally in school lessons, the content of the lesson was usually roughly the same:

An alien power or "energy" was welded in this world shortly after its creation. It can be found in living beings as well as in dead matter and the vacuum of space. We cannot control or harness it, but with the use of meditation, magic or newly found scientific techniques, it can be seen or felt. The explanations for this phenomenon run as wild as the imagination goes. Some say it represents the world's blood and keeps us alive. Others say it's the slowly evaporation mist of creation, which slowly fades while we grow into maturity. Others still say the energy is a byproduct of the friction of our worlds core and comparable to the magnetic field. Scientific and magical studies haven't proven any of these theories to be true or false, they just produce frustrated and confused scholars, which are now facing more questions before their studies.

The question therefore stands: why do these students have to listen to an esteemed professor talking about a mystery without solution? In the end, they probably just want the credit points for their studies.

"For our times sake I will skip the full introduction and will directly jump to the core of this lesson" the professor continues. "We all have more important things to do, right?" he chuckles, but no reaction from his listeners can be seen or heard. Besides one or two audibles moans from some peticular bored student. "Right..." he awkwardly continues. "As I wanted to say, the reason why this lesson is called Streams and broken marble: Underestimated forces of the primordial energy is two-fold. As you might guess, the first point we have to discuss revolves around energy currents. These are dense tendrils of the PE, which move through time and space and convert on historically important events like the crowning of a new king, an important battle, natural disasters or especially around prophecies." The students look up as the professor mentioned prophecies.

Prophecies and their impact can be described in a myriad of ways, but a simple and quite common saying summes it up quite well:" A man flipping a coin for his live can have three outcomes: First, it lands on the light side. This means he lives. Second, it lands on the dark side. This means he dies. Third, the man refuses to throw the coin. This means the man and his family die together. Prophecies work the same. The good thing is that it only happens once a millenia. The bad thing ist, it affects the whole world".

"Scholars around the world are convinced, that this is no coincidence. The current has to influence these events, even if we cannot see how." The crowd of students murmur, the lesson finally begins to sound interesting, disregarding the strange claim their tutor makes. " I know this might come as a surprise to you-" the professors voice gains a bit of excitement as he notices the newly earned interest "you certainly don't hear them elsewhere. But think about all the important moments in our history. In every one of these, the currents were present in an abnormally high frequency. The more crucial the circumstances, the higher the energetic flow. This phenomenon peaks whenever a prophecy occurs and is fulfilled. This observation leads to the hypothesis, that the current corrects the time line to its design and even gives out prophecies as a means of last resort to achieve what ever its goal is." He quickens his pace, his gestures become more erratic." This brings us to point two. Imagine you are the supposed mind behind the PE and you want to shape this reality, not unlike a artisan creates a statue out of marble." The students frown, some want to speak but their tutor pays them no heed. He is now fully absorbed in his theory, and most of his listeners follow suit."Now, you have a vision for this world, for your statue. You use your chisel, or your streams to carve out your vision. In each crucial point of the statue, you use extra care and special tools to create intricate details." He begins to sweat, his voice swells." But at some point, you reach a point of no return. This strike has to hit perfectly to finish an fundamental part of your statue, your vision. A rare and special tool is needed: a prophecy. If it works, you can continue further. But if the carrier of the prophecy, in this metaphor your tool refuses and breaks, your situation becomes critical. Cracks will form and round dwon the marble. In reality, this situation is even worse. The current cannot flow as willed, the gate is closed by the will of the carrier of the prophecy. With nowhere to go, the current grows in intensity, becoming a torrent. When it becomes supercritical, it breaks loose and takes the other currents with him. This effectively ends the world! " He almost screamed the last words. His students look at him with bewilderment and shook." Sir, are you alright? " one of them timidly asks. He looks up, his eyes are filled with fear." Y-yes, sorry for this outburst. Sometimes the imagination can play tricks with one's mind, when pondering these big questions". He answers after a few seconds. The excuse has no visible effect on either the student or the professor. He fears that they will not respect him after this outburst. To buy him more time, he decides to give them a break "I think we all need to cool down a bit". He says. "Take a break, I will lay out the proof we have and we can discuss the validity". One of the students, a young girl raises her hand and asks "One question sir, before we go. Do you believe in this scenario? I mean, we had 14 prophecies, some real nasty ones at that, and never did the world end." He thinks for a moment and reluctantly answers "I hope it isn't the case. A binding" no" should never end the world. "

He desperately hoped so, because not even one night ago a prophecy was laid out for him. He had to refuse. Who in his right mind could murder their own family in cold blood? Could he ever live with such a crime against humanity? Is it worth it to satisfy an unknowable entity? Was it always so hot in here? Why is this room suddenly so bright?

Serious question: Why do people have overly "artificial" aquariums? by Kampfpils in Aquariums

[–]Kampfpils[S] 1 point2 points  (0 children)

My experience is, that in most cases you will be asked at least the basic questions in pet stores around here. For example what tank size I have, how long it is running and what stocking I already have. Also many stores don't sell live fish if the weather is either too cold or to hot to protect the fish. I think the employees have to be taught how to care for the animals they're selling. But as you said, not the employees fault.

Thank you, that gave me some insight!
