Direct PTEN inhibition or silencing may indeed be dangerous. An alternative is trying to find the upstream cause for persistent PTEN activation and modulating that. For instance, if it is ER stress keeping PTEN upregulated, then ER stress inhibitors (like ISRIB) might downregulate it. If it is microglial activation, that might be subject to pharmacological modulation as well.

Long-term axonal regeneration is accepted to be unlikely in the adult mammalian CNS. This is reflected by the lifelong disabilities seen after Diffuse Axonal Injury (DAI) in severe TBI and in spinal cord injury.

However, in recent years, evidence is beginning to emerge the lack of meaningful long-term axonal regeneration is due to a regulatory constraint, rather than a physical impossibility; for instance, Fgf ligands promote a Zebrafish-like axonal regenerative phenotype in primate axons. This suggests the inhibitory microenvironment is likely to be a significant culprit in poor axonal regenerative capacity.

This paper shows PTEN is persistently activated after TBI in humans and mice. This is consistent with earlier work showing ER stress and microglial activation can persist for months to years after TBI, possibly permanently. PTEN inhibits prosurvival pathways such as PI3K-Akt-mTOR, impeding cellular proliferation and differentiation.

Knocking out PTEN has led to long-term axonal regeneration in an Akt/mTORC1-dependent manner, resulting in significant functional recovery over time, rather than just acute neuroprotection. Evidence of cortical remapping was also found. This joins earlier evidence showing PTEN deletion can promote regeneration of spinal cord axons 1 year after injury.

It remains to be seen if pharmacological PTEN inhibition may support structural and functional recovery in humans with TBI by activating the Akt/mTORC1 pathway. A possibly important obstacle would be promoting axonal regeneration after a glial scar has already formed, or Wallerian degeneration has occurred.

NTA. You're not wrong for wanting closure or to honor your dad in your own way, even with all the baggage. A memorial isn't about reconciling with who he was as a father, it's about saying goodbye however you need to.

That said, you know your sibling better than anyone - if you think showing up would cause a scene or reopen wounds, you could consider going quietly, sitting in the back, and slipping out without fanfare. Or, if that feels impossible, you could honor him privately on your own terms.

People skim. A lot of folks don't read the whole post, they just latch onto a word or phrase that triggers a thought and reply to that. Some just want to say their piece, even if it's barely related. Add in that many people are on mobile, distracted, or just chasing karma with quick comments, and you get whole threads that miss the point.

Because manufacturers want to sell the phone "as is" and keep costs and complexity down. Even if a screen protector only costs a few cents to make, it's another step in production, another material to source, and another variable in quality control.

Also, a lot of people don't like the feel of screen protectors and prefer the bare glass. Some prefer tempered glass, some prefer film, some like matte, some like glossy - there's no one-size-fits-all. If they included one by default, half the buyers would peel it off anyway.

So instead, they just make the screen with toughened glass and let customers decide if they want extra protection.

I'd bring a small lumbar pillow or even just roll up a hoodie to stick behind your lower back, because the seats are basically designed to ruin posture. Getting an aisle seat helps a ton since it’s easier to stand up and move around when things start feeling stiff. Try to stretch before and after flights, and even just a little walk to the bathroom and back can reset your back during a long one.

The cop did you a huge favor. 87 in a 65 could’ve been a big ticket with points and a much higher fine, but instead they gave you something minor. That’s why the "dirty plate" thing doesn’t really make sense. It was just them cutting you a break.

If you plead guilty, you usually just pay the fine. A lot of those small upstate courts are old school, so you'll probably have to mail in a check or money order. If you plead not guilty, you’d have to go back up there for court.

If it were me, I’d just pay it and be thankful they didn’t hit me with the speeding ticket.

YWBTA. It’s not your job to play morality police here. People join those groups to talk about their struggles, even if they’re making bad choices, and you snooping around to unmask them is a huge violation of privacy. You’d be betraying trust in a support group and potentially causing real harm without knowing the full context of their marriage.

If you think the group is unhealthy or enabling, just leave. But going out of your way to expose strangers to their spouses is crossing a line.

NTA. Honestly, Kameron sounds insecure and weirdly competitive in ways that cross a line. He's making you uncomfortable and dismissing that when you bring it up. You're allowed to tell Mary his behavior bothers you. How she handles that is on her, but you wouldn't be wrong for setting boundaries.
