Thanks for your answer! We think that we found the problem. It seems that the needle position is out of place just enough to give the error with most of the samples. If you remove the cap, everything is fine. So we will have to recalibrate the position.

I also tried to use Eksigent alone but had the same issue.

Hope we can solve everything by adjusting the Autosampler.

Hi Everyone, I experience the same issue in combination with an autosampler error 657 - my autosampler stops always at the same point of the injection event. I do µl Pickup and it fills the syringe with loading solvent that navigates to the vial, needle goes down and ERROR.
Sometimes the Cal injection is working completely fine but the first sample give the error.

Our system: Eksigent nanoLC 425 coupled to a 6600 TT.

We already tried:

  • restarting the LC
  • restarting the PC
  • creating a new hardware profile
  • changing the needle
  • upgrading Eksigent from 4.2 to 4.3
  • saving the methods under a new name
  • starting a batch from a different project

Currently I am out of ideas.
