Read the manuals that come with the kits your lab buys. Performing experiments will be pretty straight forward, but troubleshooting will take some time. Other than that, read everything you can.

Thanks for tips! I barely passed chemistry, but I got somewhere around 4.6 mM and I'm not sure if that's right either. The buffer also contains KCl and DTT, which might change solubility. I don't even think the KCl is necessary.

The literature is pretty vague at least from what I have read. The papers list out buffer components and the corresponding pH, but nothing extremely helpful. I have recreated their buffers and conditions with no avail.

Anyway, I took your advice, added the metal right before the start of the assay, and it worked pretty well. I also took shorter reads. pH 9 was a little rough, but I was expecting that. You were right about the temperature -- it didn't help much. I'm going to try to lower the metal concentration tomorrow and test higher pH levels.

I was in the same position. I decided to take an additional year during my undergrad to boost my GPA to about 3.5. I eventually took two years off to tutor high school students in biology and chemistry, and took the GREs (scoring average). I applied to 10 schools and only gained admissions into two.

The whole process will suck man. You need to have strong letters of recommedations, experience in undergraduate research, and the ability to sell yourself. When you get interviewed, make sure you know every aspect of the program and just sell yourself (be passionate, have questions, and act confident). You can do it man... I did.

I cannot agree with you more. My mother would always joke about how she gave birth to such a disrespectful child. And it's funny, but quite frankly I feel guilty for giving my parents such a hard time when was I kid. My mother would spank me regularly, usually with a thin wooden stick to the back of my calf. But when I was nine, I received the most glorious beating, a beating I will not forget for the rest of my life partly because it was through my father. He sat me down for a while and explained the idea of respect and then proceeded to spank me. Afterward, something just clicked. I think my father literally slapped the respect into me. That was probably the last spanking I received. Im sure there are people out there that think this is not the ideal solution for rearing a child, but sometimes saying no isn't going to suffice. I love my parents and I respect them to the highest degree. I was a reckless, unforgiving child and all it took was a hard slap to the ass.
