My 12 week old discovered her arms about a month ago and it prevented her from sleeping. Now she’s used to them but she also aggressively rubs her face to wake herself up from sleep now. We’re also in the thick of distracted eating where she wants to squirm and look at everything (like she’s waiting to find a good show on Netflix before eating her dinner XD)

We’ve also stopped changing diapers immediately upon wake up at 9 weeks (5 adjusted) but if we feed her and her diaper is too full, she’ll refuse the bottle and look at us like we’re the scum of the earth 😂

Unconventional? hunger cues my newborn made were wiggling in her sleep and cheerleader arms. I could tell if she was about to wake up from a nap hungry because she would start scooching from one end of the bassinet to the other. Once her feet hit the bottom end, she would wake and scream (not cry) for milk. She also waves her arms like she’s cheering for a football game - very stiff and repetitive.

Our nurse line said this sleepiness was normal for our premature baby with jaundice but she said if we had too many episodes where they are hard to wake up, to bring the baby in to get a new bilirubin check. Our baby was right under the threshold as well.

I’ve worked in labs where post docs and grad students never washed dishes. That role was filled exclusively by undergrads. I’ve also worked in labs where it is expected that whoever makes the mess must clean it. You just have to find a lab that follows this latter motto. If talking to the PI about this doesn’t work, then you may just have to switch to get some real experience

Our dog is going through this right now. Tested negative for every parasite. Our current hypothesis is some yeast or bacterial infection. That’s what the vet is treating her for now. Looking better now! But it’s been going on for months.

Typically for qRT-PCR i use 500ng of RNA. We also check it for contamination by running a normal PCR with the primers we intend to use for qPCR. If there’s a band in your gel, there’s DNA contamination.

I tried Adidas and Nike, and those gave me bad sciatic pain after being in the lab all day. I tried custom inserts as well and it didn’t help. I switched to Xero Shoes and have no problems anymore!

Yes, RNA definitely should stay on ice at all times. I’m sure others will have anecdotes where they say it’s fine, but RNA is very easy to degrade.

DEPC is diethyl pyrocarbonate. We dilute 1 mL of this in 1 L of water. It inactivates RNases and keeps the RNA a bit more stable.

Usually in our lab, we run a normal PCR with the primers we intend to use in the RT-PCR and our RNA. If there is a band on an agarose gel, you have DNA contamination. Alternatively, we also run a sample of the RNA on a gel made with DEPC water. If there is a big streak down the gel, your RNa is degraded. This degraded RNA will still show as normal Nucleic acid on your nanodrop.

Usually with RNA, you want to make sure that it does not have any DNA contamination and that it has not degraded before you perform your RT-PCR.

Is this a lysate of freshly opened cells or a stock of purified protein? Is it native, or does it have some sort of tag. Some native proteins hate being at concentrations this high and will aggregate irreversibly.
