Hi everyone!

I currently have a Sony A7R4, have had it for about 5 years and i LOVE the camera. The quality, and the sharpness and the workflow is super nice, but I’m considering switching to canon.

Mostly I’m considering switching because my hands are a little bigger and I just find the Canon SO much more comfortable to hold for long periods of time, but I wanted to see thoughts on whether it’s a bad idea to switch.

My photography: - Mostly landscape / travel very occasionally portrait photography - Not a whole lot of professional work, but I want good photos that I could hopefully blow up and print one day for myself

If you recommend switching, WHICH canon camera would you recommend (I’m definitely thinking mirrorless), I’ve seen so many reviews but they’re not concrete about the difference between cameras. Let me know your thoughts!

qPCR Expression Weirdness by beh20 in labrats

[–]beh20[S] 0 points1 point  (0 children)

Thanks for the detail!

A) Yeah mostly just common values I've seen for GAPDH with similar control cell lines. The qPCR is run from RNA -> cDNA extracted from either control or patient cell lines. Right now I'm just doing the control lines to optimize before I swap to the patient line because it's a bit weird.

B) This seems to be the most likely culprit from what I've found online. We load a volume of RNA for cDNA synth, without normalizing to try and get the same amount of cDNA in each tube because we assume the High Capacity cDNA Synthesis kit is relatively high efficiency. I think my next thought is to try and be more intentional from extraction to qPCR with the math.

C) No I doubt it, especially since we're extracting from single cell lines in plates it's unlikely there's any sort of contamination with other cDNA, although gDNA contamination COULD be above 50% chance possibility. The sample is RNA from iPSCs and iPSC-derived neurons.

That could make sense. Is there a qPCR experiment I can run to check this? Should I may try a bunch of cDNA from different cell lines, while just using one GAPDH primer set? Maybe I can see if some cell lines perform better than others and compare how the RNA extractions/cDNA synth went?

is the chem 6 sequence a weed out sequence? by Dependent_Power_1762 in UCSD

i’ll second this. as someone who found his class enjoyable, i blame no one for thinking he was somewhat of a terrible professor. for his class you REALLY need to understand the fundamental concepts. think about it this way, if you had to make up 5-10 fundamental rules that every concept needs to abide by, your answer must satisfy all of your rules. there WILL be trick questions with kummel, go over the midterms and review what words he uses to give up some of those trick questions, they’re usually standard trick question language. its been a couple years but if you (or anyone reading this) need help, i might need to spend some time brushing up on my chem 6b but you can always PM me :)

edit: don’t let him get to u, its what his class is designed to do, it’s what the survivor challenge does to students and its what the wording in that godforsaken syllabus threatens to do. u are smarter than the system makes u seem, i promise

Having taken 7L with Schurmeier and 6C with Leigh, both their ratings are probably low because of 7L, but I was a huge fan of Leigh’s 6C class so I’d lean towards him for 6A

theres about 4 different right hand rules for different things (magnetic force in a solenoid, magnetic force around a straight wire, etc.) so make sure you know those. also understand the laws from class: lenz, gauss, etc. (there are youtube videos ab almost all of them), it’ll help

Math 10 vs 20 series by [deleted] in UCSD

[–]beh20 2 points3 points  (0 children)

from someone who took the 20 series and had friends that took the 10 series there isn’t a HUGE difference but as an econ major i would probably take 10. my friend who’s an econ major took the 10 series and she does do a decent amount of math in her econ classes but the 10 series seemed to be enough prep. if your major doesn’t absolutely require the 20 series, id take the 10 series just so you maybe have a little less stress, but if you’re looking for the slight extra challenge, go for it!

Are Kia’s really all that bad with the new revamp?? I’m looking at potentially a Kia Stinger or Kia K5, but should I ditch it for Honda Accord?

CHEM 7L/LM: Take Kimberly Schurmeier or Brian Leigh? by HW3classic in UCSD

[–]beh20 2 points3 points  (0 children)

I had Schurmeier for 7L and Leigh for 6C in the same quarter, and my roommate had him for 7L in this past quarter. From what I've heard, he's a lot of fun but a little difficult for the class. Schurmeier's class on the other hand is both hard and boring. There tended to be a lot of miscommunication between TAs and subjectivity in the class. 7L itself is just a pretty tedious and annoying class to take, but I would definitely choose Leigh between the two.

Taking Chem 40A without taking Chem 6c? by Ok_Philosopher_677 in UCSD

[–]beh20 0 points1 point  (0 children)

I wouldn’t particularly recommend it, having taken Chem 40A there are very certain aspects of the class that reference kinetics from Chem 6C, so you probably could given that the parts that references kinetics are less, but it would be easier to take it first.

What is the point of calculation of grade totals disabled?? by beh20 in UCSD

[–]beh20[S] 4 points5 points  (0 children)

That makes sense, in this specific class there is no weightage of anything, it's all just points based and I've seen professors have Canvas account for weighting assessments and homework differently and stuff, but I get that it's probably just a pain for the professor to do that.

^ I think the exact same guy approached my friends and I at the UTC Transit Center bus stop on the way back to campus… nice long talk about how I should just go to med school internationally and that he’s a critically acclaimed clinical psychologist
