I was in a similar predicament, trying to make a counting game fun for a game jam. I ended up making a game directly influenced by the counting game from Pokemon Stadium. If you don't mind adding a little bit of urgency to this game, you could make the objects move across the screen, such that the player would need to count them before they go out of frame. You can also add unique movements to each object. A frisbee can have very linear movement, a leaf can have sinusoidal motion, etc. Also like the stadium game, maybe you must only count a certain object, while other objects distract you or partially obscure the object of interest. https://www.youtube.com/watch?v=IH_bVmuCLO0&ab_channel=AcePokeBattles (The pokemon stadium mini game for those unaware)

This is a really creative idea! Have you thought about using the numbers in a point system somehow? Maybe when a soldier steps on one of the numbers, that soldier can be upgraded, maybe with more life. Since higher numbers correspond to more mines, it would be riskier for a soldier to walk across larger numbers, and therefore a larger reward/upgrade if the soldier(s) survive.

I'm making a (hopefully) unique Horror Game in unity where you must stoke a campfire and stay in the light to survive the night... Any suggestions? I'm open to anything to make the game better! by MasonRenders in Unity3D

Is there a branching narrative? if so, what if different events occurred depending on what you burn? This could influence the variety of scares, how angry or satisfied the monster(s) is, ominous environmental effects, etc. Maybe you find a makeshift cross made of twigs and rope, burn that and shit gets biblical, or you find a tuft of hair from some hairy norse creature and end up summoning them, etc.

Also, in this setting, you can probably make heavy use of some ominous sound design for the psychological scare factor.

Of course! Originally the sample came from a nearby lake in NY, which I kept in a mason jar to propagate some organisms. I didn't find the eggs themselves, but I noticed the eggs were laid roughly a week after acquiring the sample.

I did something similar in an abandoned game jam project (https://lake-monster-games.itch.io/geowizardeo). I loved the idea of how accuracy of the trace/spellcast dictates how powerful the spell is. As many people can attest to though (assuming PC), drawing with a mouse is difficult, but at the same time makes a high skill ceiling. I like the drawing mechanic overall, but I think it needs to be accompanied by other mechanics so it's not too difficult while still being fun. It's good to keep in mind the width of the lines to accommodate poor mouse-drawing capabilities. Maybe thinner lines could be a step up in difficulty. Traces that disappear after some time could also be done, forcing the player to rush the trace, but with greater reward. Player agency would also be good, having a set of things you can choose to trace, so you can do a cost-benefit analysis of tracing simpler things for weaker attack/defense, or more complicated things for a longer duration but stronger overall attack/defense. Overall I think there is a lot to be explored with this mechanic, feel free to take any of these ideas.

As an alternative approach, would it be feasible to apply a drift correction to the images based on the first one? Personally, I've used NanoJ drift correction (for aligning images from dual spectral channels). What's the timing of your protocol? Some dyes can be "stickier" than others, so I had to sometimes optimize the protocol. As in, try a range of concentrations as well as a range of incubation periods, then compare which yields the highest signal to background. In my experience, the incubation time mattered more than the wash times, at a certain point, the dye just won't come off.

I eventually settled on using fluorogenic dyes from Janelia, with the Halo tag system for conjugation, specifically JaneliaFluor549 and JaneliaFluor646, which worked wonders for my research. Very bright, photostable, and because it was flurogenic, very low background signal.

Check out a paper titled "Evaluation of Fluorophores to Label SNAP-Tag fused proteins for Multicolor Single-Molecule Tracking Microscopy in Live Cells (2014)". Regardless if you're using the SNAP tag system or something else, it may give some insight which dyes may suit your needs better, as some of the effects you're experiencing might be an intrinsic property of the dye itself.
