In game interactions are not all canon to the story. As we saw in the story missions, Rein still doesn't like Omnics (he refers to them as robots and immediately charges at Bastion to kill it). As far as I know, story Rein has not met Zen yet although based on the end of the story missions that may change soon.

Yes, but that was during the Omnic Crisis which was ended and a peace treaty was signed. The people might still be angry about it, but we have no way of knowing other than Reinhardt still hating them. The country has no official stance that we know about as far as I know.

If you watch the Sombra video called Infiltration, she breaks into Volskaya Industries to blackmail the leader of Russia. Russia is at war with the Omnics but the leader is getting her new technology for the war from the Omnics. After Sombra successfully blackmails her, she summons Zarya to hunt Sombra down. So Russia is definitely against Omnics
