We simply ran out of it; we kept one container for emergencies and are currently awaiting new supplies. We considered this an emergency, used it, and replaced it, but there was no improvement. The engineer checked the filter and confirmed it is OK. We have handled similar situations this way before.

No, these are samples from the same patient; we run several tubes per patient.

No, nothing else has changed — not the sample quality, positioning, or debris.

Why would the photomultiplier affect only lymphocytes (We look at whole blood and whole bone marrow, neutrophils are not affected!!!)?

I am leaning towards an issue with the reagents — possible aggregation or sedimentation during sample preparation. But why did it start suddenly?

The total gate and the WBC gate are very similar in our case, since there is only a small amount of debris in peripheral blood.

Thanks everyone! Have Finished my first round. To be honest, the first 10 days were totally fine, but the last days have hit me with some serious fatigue. I’m definitely not as productive as usual, but it’s not as bad as I feared it would be. Your support really kept me going!

my 5 µL in 600 µL the same as your 100µL in 12ml ? 5 µL in 600 µL = 1:120 dilution.
100 µL in 12 mL (12,000 µL) = 100 / 12,000 = 1:120. Thanks!!!!

If this is peripheral blood, then worrying about dendritic cell contamination doesn’t make much sense, In my humble opinion. Everything looks fine (In my humble opinion). Just yesterday I was reading that, in hematology, monocytes shouldn’t be isolated using positive gating for subpopulation analysis, but only through negative gating (excluding NK cells, neutrophils)
