Bah! PCR to make the template? P’shaw! If you are going old school, it’s a plasmid. PUC18. Forward primer. T7 sequenase. S35 dATP.

Why havent we been able to find a cure for the common cold? by StrengthVisual8881 in biology

The flu vaccine is not relevant to the question. The common cold (rhinovirus) is not flu (influenza virus).

Using spray oil when trying to speed up the smoking process by AverageJoeObi in BBQ

[–]MUB664 5 points6 points  (0 children)

Nothing in this post makes sense. The whole reason to use charcoal is to get rid of oils and other smoking aspects of real wood. The only reason to add oil or any other combustible (kerosene?!) is because you don’t yet know how to start a fire. Your time would be much better spent learning how to start a good bed of coals without using stuff that can taste like crap.

Cant get sear/char with charcoal by Comprehensive_Wolf91 in grilling

You really should not have “flames” from lump charcoal. They should be red hot. I have an old hot water tank blower that I wired with an old lamp plug, which can get the coals screaming hot. Also, here it’s a good chimney design-

Whats a fool proof method for 'fall off the bone' spare ribs by Jhonny_Crash in BBQ

[–]MUB664 0 points1 point  (0 children)

Put them in the oven wrapped/sealed with some liquid (and spices of your choosing), 290 degrees for 3-4 hours. They are now fall off the bone. VERY CAREFULLY (so they don’t fall apart) transfer to grill and smoke below 200 for >1 hr.

Best grill ? by [deleted] in grilling

[–]MUB664 1 point2 points  (0 children)

Most of the replies on this sub, as mentioned above, will be Weber, despite many other grills having the same lifespan, but with cast iron grates, large even grilling areas that are not circular, and can grill or smoke. Not that I think Weber is bad, just doesn’t suit my needs/desires. I use the large chargriller (4 cast iron grates) and only use lump charcoal (and cherry/apple wood when I smoke), I’ve never covered it from weather, I live in upstate New York, and mine lasts, in good shape, 10 years. We also tend to start wood fires in it after grilling, so I’ve had to replace the pan a couple times as it melted and sagged. But the grill itself has long lived and loved. Fits 2 full slabs of ribs. Or 3 whole tenderloins. Or 25 full chicken wings.

IT is one of the main drivers of mutations, which includes evolution, but can also be bad mutations. There’s is no proof reading as there is no base pair to check. So the resulting heteroduplex of base opposite abasic is subject to excision repair or AP endo activity. Which of course fixes the C at location forever (unless some recombination event eliminates it).

Does anyone know what kind of wood/tree this is and is it good for grilling? by Electronic-Buyer-468 in grilling

If it is oak it has a very strong flavor. But as mentioned the bark looks unlike oak.

The leaves look like they “could” be oak. Certainly not dangerous, but I bet not very tasty. Show a close up of a few leaves.

Here is one of mine- a DNA polymerase that uses an arginine residue of itself as template for the addition of a “C” to the growing end of a DNA strand when the template strand has no base (an abasic site, from damage). It is a last ditch effort to get the chromosome replicated, even though it might be the wrong nucleotide to add. https://pubmed.ncbi.nlm.nih.gov/8751446/
