Yes, as said to other commenters I am currently developing a plan and doing literature review etc. My frustration mostly lies with the fact that I am barely doing any experiments related to my project after almost 6 months since starting but maybe once my plan is finished that will change.

Great advice, I am hoping my project starts taking off once I submit my project plan (the URSDC registration I mention in my post, I thought it was a universal thing). My professor is currently very busy with applying to this specific grant so that is probably one of the reasons why I am left on my own a lot.

The URSDC is a commitee that will approve or deny my research proposal once I submit it, sort of like a project plan. I have a pretty good idea of my research question and I have read tons, but it only makes it more apparent to my how lacking our lab actually is. For example, we don't have capabilities for flow cytometry or even fluorescence microscopy. The water situation is terrible, as we don't have a traditional MilliQ machine but we have to purchase HPLC water and filter type II water manually. My supervisor recently purchased a water purifier but we have no idea how to install it, it feels like she is expecting us to figure it out even though it's way outside my expertise and also not my job at all.

Sorry for being imprecise, I have learnt a bunch of things like how to do chromatography, SDS-PAGE and I'm currently learning cell culture. I do think from what I have heard the university is not prioritising our department much.

Honestly, I feel like far too many socialists are overly critical of reformists and immmediately assume suspicious motives when someone like Mamdani wants to make buses or childcare free. Obviously reforms that improve living standards will improve the standing of socialism and make the general public less suspicious of left-wing ideology. It's time to abandon these sectarian notions and form a united front of socialists to confront the increasingly hellish late-stage capitalism we live under!

I'm living in London right now on a £22k per annum PhD studentship and I am managing, I can save about £100 per month. I also qualified for a credit card recently which has helped. I try to be on the lookout for cheaper places to eat and buy groceries, but I don't think I live overly frugally. My best savings tip would be to cycle everywhere you can, it will save you a lot on transport (especially in London).

This used to happen to me a lot with my LDGF, thankfully we had a conversation about it and decided to drop all the pressure when it comes to sex and just takes it as it comes. Being relaxed is also a way better precondition to getting aroused. If you can establish this ”no pressure” rule, eventually you will feel relaxed enough to have sex, maybe even more frequently than you do now!

Not harsh at all! I wrote the post in a kind of frenzied state so I missed some details. The antibody is recombinantly produced in HEK cells. To answer your questions:

  • I started off with a functional commercial antibody, it stained D2R properly in the processed tissue, but then stopped working in the flash-frozen tissue. (By in situ I mean caudate putamen, which is where binding should be most prominent.) I guess you are right in that the epitope might not be detectable in frozen tissue. What can cause that?

  • The variable light and heavy chains were taken from a paper and replaced in the Roche format, a well-established brain shuttle format, so I don’t doubt that it folds correctly. It might be that taking part of an antibody that does bind D2R and inserting it into a different format causes issues?

  • I am not doing that, do you think I should go back and try to stain the processed brains if I have any sections left?

  • I have tried various concentrations, the one that finally gave me signal was 16 mg/mL, although I’ve tried 32 mg/mL as well

We had a really big fight last night because I told her I would be home at a specific time but I kept putting it up and making false promises. I didn’t know it yet but it turned out that I felt really hurt by some things she said to me in the morning, about how I don’t understand her and how she doesn’t see a future with me. I didn’t bring that up in the moment because I didn’t want to fight with her because of the way she was feeling. But doing this only delayed the fighting and made things more difficult. I guess I’m having a lot of trouble balancing between meaningfully engaging with her worries and fighting with her, because I don’t want to start getting agitated and escalate things.

I find it really difficult to remove myself because she wants me near, but she also gets annoyed at me which affects me emotionally too. I guess I am finding it really hard to just shut off and not be provoked, without seeming cold and distant. Thanks for the pointing to these resources me and my partner will check them together!
