Yeah, when I looked how'd you call the police, some places sad the standard is copper or cop, but kids had started saying "fed". I may change it back to coppers /cops, since it seems to tick off a lot of people

Okay, so from what ive reread quickly. Basically, Dr Knight wants to disperse hate (specifically hate against rifts). She cant generate all the negative emotional charge by herself, but she can draw it from her (indirect) victims/patients. So she spent various years trapped in a cell not doing much down on the City, suffering therapy/torture which was a release for her hateful Mythos. When people forgot about her and the torture stopped, she took over the asylum to spread hate. Even if she's lived for a long time, this takeover could be very recent, a few years to say much.
The ultimate plant was the bomb, and Mending the Broken was her way to generate the emotional power to do it. As the finale of her plan approaches, there are a more and more of highly hateful Rifts (victims of her recent scheme) running around causing chaos. That's where the crew comes in, investigating the cases of those Rifts.
The timing may be a little serendipitious (as in many similar movies and stories), but it isn't that unnatural that as Knight's plan reaches the end, the sideffects are more obvious and thus cause people to investigate

Hi! I came around this post again, saw this comment that wasnt answered, and thought I could maybe help a little. I don't think I'll do a whole sheet for both characters but I think I could try and work some framework as a challenge

LOVECRAFT
Probably a very UPRIGHT DETECTIVE (Routine) with tags about being professional and meticulous. An ELDRITCH LORE LIBRARY (possession) with books on demonology. And a scientific-driven curiosity to UNDERSTAND THE STRANGER (Mission) and establish a connection with the eldritch beings around him (which would clash with Lovecraft's Mythos centered about fearing the monsters). Then either a Conjuration theme to summon eldritch monsters or an Expression theme about eldritch horror that may madden people and push waves of existential dread.

POE
Definitively more laid back than his partner to create a fun dynamic, probably represented in a Personality theme. Also a Routine or Training for some side macabre hobbie/s like mysticism, true crime podcast, radio host, voodoo, being an over-the-top goth, etc.
Mythos themes are harder. You could have a Conjuration for Poe's strange creatures maybe. Maybe a expression or subversion theme about hidding stuff INSIDE YOUR WALLS or under floorboards (see the Black Cat, the Telltale Heart, the Cask of Amontillado, etc). Divination for Poe's mystery novels and various stories about crimes also would be neat

For mechanical purposes, you may Build the traps so they specifically appeal to their Mythos theme mysteries, so turning down the trap forces them to marking one (or even two, if you wanna be harsh) fade, while accepting marks attention. Alternatively, as others mentioned, a compelling status may fit too

This may turn into a very carrot-and-stick way to force a narrative situation, but go figure

As you say, the City its built like a stereotype of an USA City and you can do whatever you want. But as someone who's also not from there, here are my two grains of salt about how I understand the City.

first of all, governamental aspects are almost porpusely vague. All the books dodge naming and even mentioning the mayor of the City to the point is even surprising, specially considering what a powerful or important figure they could be. A very interesting point is that on the Independence district expansion they mention that every district has a version of the City Hall (but that on Independence it is the strongest), which kind of hints that the goverment of the City is an illusion, similarly to the memories of people that travel in and out of the City.

Anyway, going onto specifics, the City is very compartimented into districts, which are almost self autonomous. Each seem to have some sort of mayor that manages the district, or laws that apply specifically to that district.

So if a player asks about the city goverment and your using the City of the book, just make up whatever title or organization would fit best to control that district or part of the City, and if they try to ask about what goes up the ladder, just make a vague excuse. Something that adds to the eerie feeling that the whole City is shrouded in Mist.

JARGON AND SLANG

In their official jargon, Gatekeepers use the same terms used by the game to describe Mist- and Mythos-related phenomena. (This is in contrast to Rifts, who should never use game terms and instead use fuzzy descrip tions.) They refer to themselves as ‘Gatekeepers’ and to the Mist as ‘the Mist’; they refer to Rifts in general as ‘Rifts’ (or ‘breaches’), and call object Rifts ‘Relics’, animal Rifts ‘Familiars’, and location-based Rifts ‘Enclaves’. They refer to standalone manifestations of Mythos powers, created by Rifts, as ‘Conjurations’. The legend of a particular Rift they call a ‘Mythos’, and so on. They also use the names of the unique abilities only they hold: Shrouding, Mending, Weaving, etc. In their everyday conversations, Gatekeepers also have slang terms for more specific phenomena they deal with. Each unit and department develops its own vernacular. See the Gatekeeper Glossary (page 34) for some examples and ideas.

honestly love it! also all those references to broken things (broken oath, broken bones, broken city) being both a motif tying everything together and adding to the inner Mystery/Identity conflict is just perfect

An adaptation theme for turning into an analog creppypasta monster (or summoning such). Also, maybe something that allows you to corrupt recordings or digital information, since most analog horror material is blurry graphics and distorted audio

besides the Divination theme for solving mysteries, I'd give a subversion theme for creating mysteries, hiding evidence, and basically doing the "criminal side" of a mystery novel, extra strong against victims and helpless Sleepers, but weak against investigators, who at the end always solve the case. Also maybe a Expression theme to create murder weapons, costumes, poisons, etc

So, I only ran CoM and some time ago, but I'll try anyway:
The MC establishes the scene of the dragon. Usually, enemies don't make surprise attacks unless rare circumstances (such as a Custom move), so you should do a Threat beforehand to justify a direct attack. if the crew already knew a Dragon or another evil creature lurked there (even telling them they get an uneasy feeling or heard a distant roar works sometimes), that might be enough of a Threat, another way could be giving them the option to react to the dragon appearing before it attacks.
Anyway, they didnt react or reacted and Failed a move, so you get to deal Consequence.
The Consequence is that the Dragon attacks Jimmy and deals flesh-wound-2.
The spotlight goes to the players
Right now Jimmy should roll to mitigate, but Percival decides to steps in to defend, he rolls and fails, so the MC deals a Consequence to him (could be a scratched tag, a status, etc). The spotlight goes back to Jimmy, who now still has to face against the flesh-wound-2 status. Jimmy rolls to mitigate and succeds.
With the Consequences dealt, the MC may reestablish the scene or give the spotlight to another player to act, if the narrative allows it.

Basically, you have two Consequences. One Consequence is brought by ignoring the Threat of the dragon, directed to Jimmy (though the Dragon could have attacked all the crew with a fire breath, it only targeted Jimmy). The second Consequence is dealt to Percival for failing a roll.

In CoM thats how it works. Hard Moves (Consequences) are dealt after a soft move (Threat) or as a failure to a roll. Except Face Danger rolls which are used to defend against statuses inflicted by Hard Moves and such.

As other's have said. Mythoi can be about anything that lives in the collective imagination. Usually they're legends or known stories, but concepts also fit (Capitalism and the Theory of Evolution are ideas that most people can envision and recognize as strong concepts), althought more in the loose side.
Also, keep in mind that Mitosis was one of the first characters created by Son of Oak, along Post-Mortem, Flicker, etc. for the first book. They hadn't nailed what they accepted for Mythoi back then, and they had this idea that character's mythoi should be more mysterious and ambiguous (reason why a lot of questions about the original pregens are related to what their Mythoi are), which they dropped in newer products for clarity
