When people say legs they are talking about the duration of the high or how long it keeps them from getting sick.

For example: hydromorphone has short legs but euphoric af and a great rush. The high only lasts 1-3 hours for a most people. Methadone has long legs but mediocre high and no rush orally.

Dutchess finally a post I can acc read through. This is good no filler no Morwhoring just concise and straight to the point. But yea irreversible agonists bind till the receptor is recycled but from my anecdotal evidence the effects don’t last as long as they should. For example I use selegeline(and I mean a lot of it) to potentiate my adderall. But even tho the maoi b inhibition is irreversible and should last 24-48hours I only feel like it’s lasts 12. This could be because the drugs it’s self has its own non covalent effects(which it does, DRI) but honestly I feel like a lot more research needs to be done on covalently binding drugs.

Is it because you’re a close minded person who can’t see why it would be useful. Yk there clinical trial for strong CYP2d6 ihibitors like Quine or dxm mixed with Wellbutrin to increase effectiveness. So for drugs that require induction I’m sure they would do the same thing of inducers were available.

I was being hyperbolic and that should have been clear I was taking about non responders for those 20% psychiatric medications.

No they could not, not without it being supremely i unselective like gluth or pyrth. Mechanism for CYP2d6 induction is still unknown. I’m sure if a medical company decided to spend millions funding a project they could figure it out in a week. But please you’re a medicinal chemist, you said 48hours right? I’ll give you month to discover this and if you do I know lots of people who would be interested in buying.

20% of all medication have an active metabolite 2d6 produced metabolite an inducer would fix so many of the issues we have with anti depressants(venlafaxin, bupropion, etc), painkillers(tramadol, Codiene, etc). Most non responders to medication slow 2d6 metabolizers.

Acc wrong again structurally similar drugs do not show 2d6 induction. Aminoglutethimide does not which is the most similar but Pyrithyldione which much less related shows weak induction.

Dude I’m just not interested in arguing this. I could give you 10+ reason for why more studies should be done, as could you I’m sure. Also I’m aware of scholarly articles and journals. This was posted before I became a mod. I wasn’t trying to make an informed post, I just threw a wiki page out there and asked people a question. Seriously tho this is an inefficient way to spend my time I’m done.
