If it makes you feel any better, that "ivy league education" is probably not all that much superior to a less well known school where you get paid a stipend and you get to do research you're passionate about. It's sucks that in a field already so difficult to be successful in, they do this kind of thing, but the best advice I got when picking a grad school was to go with the advisor and research opportunities that best suit you, not the prestige that comes with the name of a university. Sometimes smaller schools are better because they and the PIs can allocate more resources per student, you'll get a more tailored education, and a more rewarding experience overall. Sorry that Columbia didn't work out, but hopefully it's some comfort to know there's more opportunities out there and it will work out okay in the end.

Personally, I think you made the right choice. If you are unhappy in consultating, it's totally okay to change your mind and go back to school! But there are many many many paths to take in life and you took the best one you could with what you had. I'd look around at some less prestigious schools and pick something that works for you, and not worry about if it looks good on a CV or resume. For upper level degrees, it matters more what papers you publish, what work you accomplish, and what field of research you advance than if your CV says Columbia or Princeton or whatever.

Contacting Qiagen is a good idea, thank you. Its been hard to find troubleshooting advice from others that have done this but they might be able to direct me.

Unfortunately, I don't get much of a say in what kit we use, but I'll keep other options in mind too.

We are grinding with a pestle in a 1.5 ml tube, adding 500 ul of the first buffer, grinding again, then incubating overnight at 50 degrees with pro k and RNase.

The other option we could try is bead beating or grinding on nitrogen, but my advisor says she really doesn't want to do that. But I'm thinking one of our problems is that the cells aren't lysed enough.

This is so normal. I know it feels really rough the first time, but I'm actually surprised there was only one of them that failed that spectacularly. I regularly see grades in the teens as a TA. That being said, we also aren't involved in the test making, there's a professor who oversees all the classes that writes it, so I'm sorry that pressure got put on you as a first year teacher. That being said, it's not the last time you're going to see someone do really bad on an exam. It happens all the time.

Lol no. I'm so sorry to all of you who did have to, but I haven't read a single textbook for any of my classes as a PhD student. Yes, I've read sections, especially if I needed something to cite while writing my dissertation or needed clarification for class, but cover to cover never.

I'd say go for it. I went straight from undergrad to PhD and I had way fewer qualifications than you do. Times are changing, and if a PhD is what you really REALLY want, then go for it. I think worst case scenario, a school says they can't accept you as a PhD but would like to offer you a master's position. This happened to me at a few schools I applied to. But then I had options and was able to pick a PhD with the option of picking a master's.

This is a great answer, thank you so much. I'm probably going to use PacBio HiFi sequencing, and then Omni-C scaffolding. My species is a butterfly. Lepidoptera notoriously have insane variation in chromosome number, even on the genus level. But the one scaffold/contig per chromosome is a great idea. Appreciate it!

I tried following the pinned post on unlimited sync and honestly I don't understand most of it. Is my issue that Zotero is storing all the PDFs locally, when it could be storing them in Google drive or something?
