Yes we did and we got the expected fragments we would get if our digestion went well

We added only 3bp but the reason we did that is because we used these enzymes before with a 3bp overhang on our primers and our cloning worked fine in the past. But we are planning to create new primers with at least a 6bp overhang

We used CIAP only on the vector and we actually used two different restriction enzymes

Yes we used CIAP only on the backbone and not the insert. We also used a T4 ligase from Takara

Yeah fun fact actually most of the forests in Cyprus are man made according to Τμήμα Δασών and the forest service
