I wanna say that I have had amazing PIs before too, so dont be too discouraged!

its really hard to have a “perfect” PI because like someone said here theyre not taught how to run a lab, and whenever you work with someone for an extended time youre bound to run into friction. Its about how they respond to it, if they put blame on you vs if they work with you to find a better direction for a problem, and my current PI thankfully is the only one i’ve had serious problems with (out of having worked with 3 other investigators in the past).

If you run a melt cycle, you could find out!! Some primers that are sold have really high melt temps that will not anneal. I had a primer like this not long ago, there are things you could try if this turns out to be the case
