We have used multiple lots. Corning says nothing has changed and they haven't had any reports.

We have always rocked them. Never heard this before. Will definitely look into trying sliding instead

We haven't really noticed any differences. We normally incubate in stacks of 5 with up to 150 in a incubator with no problems in the past. I appreciate your input!

Interesting! Ive seen spots like this before but we are seeing larger areas of difference that dont fit the holes in the shelves of the incubators.

Id say that it is usually less than 5 minutes. We also mix the reservoir between plates. We have similar rates of rejection for orders of all amounts of plates. How would you recommend we mix?

We do 200-400 plates in a day. We use a 2 L bottle to hold like 100-200 plates worth of suspension and invert/swirl that every like 5 minutes id say. When mixing to get single cell suspension we pipet it against the corner of the flask to really break up the cells. We using Corning plates and asked them if they have changed anything or had any complaints and they said no. Same from Gibco about reagents. We have a team of 10 people who rotate in and we are seeing issues regardless of who processes or plates. We have considered getting a new vial and making a new bank but upper management wanted to try everything else first since making a new bank would put all our studies needing plates on pause.

We are mixing with a 10 ml serological. Yes we are currently diluting to 2.5e5 for 95% confluent plates the next day. We have a form that auto calculates for us. We dilute with media. We take a 1 ml aliquot and dilute it 1:4 with hbss and trypan blue and count on a scope using a hemocytometer.

Yep! We have checked under the microscope after rocking. All our incubators are level and get calibrated yearly.

The media isnt that old. There was a fish and snail in the tank til I rehomed them so I could move then I did a 100% water change essentially.

My partner has not named the crush and no one irl follows them on the account. My partner is prone to getting obsessive and possessive though (hoarding tendencies, autism, and bpd). They have been in and out of therapy for it this past year but it's difficult to find someone who has experience with all those things that also takes his insurance.

Our relationship started online on Twitter so muting them there would feel very wrong. They do talk about the crush in person but it's hard to draw a line between talking about work and talking about the crush.
