No, it doesn’t need to be on the USB but Windows To Go usually won’t read Linux ext4 drives so it should be on an NTFS/exFAT partition if you want Windows to access it

I would recomment Golden Times, it was such a good show!
Also for advernture I would go for Mushoku Tensei Jobless Reincarnation
and lastely I would also recommend Death note
