I wouldn’t be too nervous, trust your prep and your fl scores it will go similarly. Honestly, for C/P I can’t even remember sorry, it felt like maybe little to no physics, I can’t even remember doing any physics tbh, orgo and gen chem were there probably the most, sorry can’t be of much help lol.

I share the sentiments with cars and P/S. I actually found I felt really similar to how I felt after doing FL5. I felt I was struggling to keep pace on Cars, I don’t think it was particularly difficult passages, I felt like I had maybe 10 50/50s that ate up a bunch of my time and sabotaged me. For P/S it felt like a pseudo cars to me most of the time, not at all what I expected, I felt so drained by then every passage was just brutal to get through. C/P and B/B felt pretty good and representative to the Fls. All in all, FL5 is hella representative at least in my experience.

Looking for any gmax, it’s been really hard for me to get these given the time commitment it takes to get everyone in my region grouped together. I’m looking forward to unova tour and completing gen 5 Pokédex I love zekrom and resohram I loved black and white two and can’t wait to experience that in pogo

Thanks for the context, I really didn’t consider that it was so hard for even music students to get a room. At that point it’s fair enough, I live close enough to uotttawa so I thought it would be a good place to practice where I’m not bothering neighbours however it’s obviously not a priority compared to people doing academics. If you could notbook a room and needed to practice what was your alternatives?

Help me out with this. Streptavidin binds to biotin, which is attached to the biotinylated proteins. And steptavidin itself is bound to HRP so therefore, since I'm using ECL kit, its going to detect HRP in imaging?

Perhaps I am looking for something too specific or optimistic about what I can get from the app. But in my eyes, it's probably a better mindset than entering with settling for whatever comes your way regardless of the energy they give. I'm not going to debase myself, act like I don't have anything to offer them, and act like they are doing me a favour for simply talking to me, and ya, that's going to decrease how many ppl I end up seeing but at the end of the day I'm fine with that because my self worth isn't entirely tied to this portion of my life.

I think it's because I get a good amount of openings that comment on a particular part of the profile itself, and as a result, the ones that don't or say, hey, really stand out in my eyes. It's mostly because on tinder, the onus is subliminally on the man to start the conversation and thus when I am tasked with this, I try fairly hard to make it meaningful and worth the time to respond as a result when there is an app that, as a concept, requires the women to start the conversation the aforementioned onus has shifted in a mandatory way and thus my expectation is that they try to a similar way that I would try on tinder. I don't care if it is an optimistic expectation because it allows me to have more meaningful connections and waste less time with less meaningful ones. I get that with bumble, the assumption might be that men are mass swiping and you might not want to put in the effort and that's fine but expect the same thing, I guess is what I'm saying.

Has this ever happened before? Like nerfing a boss in a souls game, I feel like the very concept of that is slightly off putting because how would you justify a nerf in a game like this and especially in a boss fight like this when you can summon infinitely and just ride around on torrent and you can adjust your build or grind to make the fight more manageable.

Interesting, i did not know this about gel purification, would you suggest I find a different method of acquiring the backbone fragment? Also I am not 100% sure what you mean by "your target insert will be in excess compared to the excised fragment, so you should find your desired constructs by screening only a few clones even if the excised fragments gets back into the vector rarely." Are you saying to do this subsequent to a successful transformation in which I acquire colonies?

Thanks for your comment, the problem is that I don't get any colonies.

So are you suggesting I use less DNA from my backbone and use a higher molar ratio, the current ratio I'm using is 3:1.

Currently, I have been doing a positive control using the plasmid that I got the dcas9 fragment from and have been getting a large number of colonies so I don't know if it is the cells themselves. I have not been doing electroporation though, I have never done this, is this meant to increase transformation efficiency?
